摘 要:齐口裂腹鱼Schizothorax prenanti是我国重要的冷水鱼类之一,为我国长江上游名贵经济鱼类.该研究在免疫组和对照组齐口裂腹鱼胸鳍基部分别注射0.2mL体积单位为1.0×108 CFU/mL福尔马林灭活的嗜水气单胞菌苗(F-AH)和0.65%灭菌生理盐水,在免疫后第1,2,4,7,14,21,28天,测定外周血液血细胞数量及组成比例,血细胞吞噬活性和血清抗体效价.结果显示注射灭活菌苗后红细胞、白细胞、淋巴细胞、中性粒细胞和单核细胞的数量逐渐增加,显著高于对照组;血栓细胞的数量逐渐降低,在第4天和第7天显著低于对照.吞噬百分比(PP)和吞噬指数(PI)迅速上升,第2天达峰值,PP第2,4天显著或极显著高于对照组;PI第2,4和7天极显著高于对照组.血清抗体效价逐渐升高且在第7天和第14天达到最高值.结果表明单次注射灭活菌苗后齐口裂腹鱼可以产生良好的免疫应答.Prenant’s schizothoracin,Schizothorax prenanti(Tchang),is one of the most important cold fish species in China,and it is an important commercial fish distributed in the upper reaches of the Yangtze River.In this study,each individual of S.prenanti was injected at the pectoral fin base with 0.2mL of the bacterial suspension of formalin-killed Aeromonas hydrophila at a concentration of 1.0×108 CFU/mL for the immunized group,and with 0.2mL of 0.65%sterile saline solution for the control group.Blood samples were taken from each fish of the two groups 1,2,4,7,14,21and 28days after injection,and the haematological indices,such as leucocyte and erythrocyte number per microliter and differential leucocyte count(DLC)of the peripheral blood,activity of phagocytes and serum antibody titer were determined. The number of leucocytes,erythrocytes,leukomonocytes,neutrophils and monocytes in the peripheral blood of the immunized group drastically increased,and was significantly higher than that of the control group,and the number of thrombocytes in it gradually decreased,and was significantly lower than that of the control group on day 4and day 7.The phagocytic percentage(PP)and phagocytic index(PI)also dramatically increased,and peaked on day 2.PP of the immunized group was significantly or highly significantly higher than that of the control group on day 2and day 4,while PI was highly significantly higher than that of the control group on day 2and day 7.The serum antibody titer gradually increased and reached the highest value on day 7and day 14.It is concluded from what has been stated above that the resistance of immunized Schizothorax prenanti against bacterial pathogens was enhanced by means of increment of nonspecific cellular immunity.
