摘 要:研究铬暴露对草鱼Ctenopharyngodon idellus鳃抗氧化酶活性和脂质过氧化作用的影响.将草鱼暴露于不同体积质量分数梯度(0mg/L,7.23mg/L,14.47mg/L和28.94mg/L)的六价铬水体中,分别于24h,48h,72h和96h测定鳃中总超氧化物歧化酶(T-SOD)、谷胱甘肽过氧化物酶(GPX)的活性和丙二醛(MDA)质量分数.结果显示在24h时,随着Cr6+体积质量分数的增加,草鱼鳃的T-SOD活性无显著变化;在48h,72h和96h时,随着Cr6+体积质量分数的增加,草鱼鳃的T-SOD活性表现出先升高后降低的趋势.在24h,48h,72h和96h时,随着Cr6+体积质量分数的增加,草鱼鳃的GPX活性均表现为先升高后降低的趋势,24h时在14.47mg/L组达到峰值,48h,72h和96h时,均在7.23mg/L组达到峰值.在24h,48h,72h和96h时,MDA质量分数均随着Cr6+体积质量分数的增加而增加.这些结果表明六价铬可以引起草鱼鳃组织发生氧化损伤而使其出现中毒现象,且抗氧化酶活性与MDA质量分数之间可能存在一定的负相关性,表明MDA质量分数增加可能与抗氧化酶活性的下降有关.To study the effect of chromium on antioxidant enzyme activity and lipid peroxidation in the gill of grass carp(Ctenopharyngodon idellus),grass carp were exposed to Cr6+at 0,7.23,14.47or 28.94mg/L for 24, 48,72or 96hours,and then the content of malondialdehyde(MDA),and the activities of glutathione peroxidase(GPX)and total superoxide dismutase(T-SOD)from the gill of the treated C.idellus were measured.The results indicated that in the 24-hour treatment,Cr6+concentration had no significant influence on T-SOD activity,while in the 48-,72-and 96-hour treatments,the activity of T-SOD increased at first and then decreased with increasing Cr6+concentration.In the 24-,48-,72-and 96-hour treatments,the activity of GPX increased at first and then decreased with increasing Cr6+concentration,and its maximum occurred in the 14.47mg/L group in the 24-hour treatment and in the 7.23mg/L group in the 48-,72-and 96-hour treatments.MDA content increased with Cr6+concentration in the 24-,48-, 72-and 96-hour treatments.The above results indicated that hexavalent chromium can cause oxidative damage of grass carp gill and make it poisoned,and that a certain negative correlation may exist between antioxidant enzyme activity and MDA content,thus suggesting that the increasing of MDA content may be associated with the decline of antioxidant enzyme activity.
