摘 要:目的研究过量氟在大鼠体内的蓄积以及钙的拮抗作用。方法将80只初断乳清洁级SD大鼠随机分为8组,分别为对照(蒸馏水)组、单独加钙(100 mg/kg碳酸钙)组和低剂量(2.1 mg/kg Na F)、中剂量(8.5 mg/kg Na F)、高剂量(34 mg/kg Na F)单独染氟组及低剂量(2.1 mg/kg Na F+25 mg/kg碳酸钙)、中剂量(8.5 mg/kg Na F+50 mg/kg碳酸钙)、高剂量(34 mg/kg Na F+100 mg/kg碳酸钙)钙氟联合染毒组,每组10只,雌雄各半。采用灌胃方式进行染毒,每天1次,连续染毒90 d。每15 d测定24 h尿氟浓度,染毒结束后测定骨氟含量。结果与对照组比较,低、中、高剂量单独染氟组和中、高剂量钙氟联合染毒组大鼠骨氟含量升高,低、中、高剂量单独染氟组和低、中、高剂量钙氟联合染毒组大鼠尿氟浓度升高,差异有统计学意义(P<0.05)。随着氟暴露剂量的升高,大鼠的骨氟含量和尿氟浓度均呈上升趋势。与相同剂量单独染氟组比较,钙氟联合染毒组大鼠骨氟含量有所降低,差异多有统计学意义(P<0.05);而不同性别大鼠在不同钙氟联合染毒组、不同时间点下,钙对尿氟浓度的拮抗作用程度不尽相同。结论适量的钙可拮抗氟在大鼠体内的蓄积作用。Objective To investigate the accumulation effect of fluoride in rats and the potential antagonism against these effects by calcium. Methods Eighty healthy SD rats were randomly divided into eight groups: control group(distilled water),calcium control group(Ca CO3100 mg/kg),low-dose(Na F 2.1 mg/kg), middle-dose(Na F 8.5 mg/kg), high-dose(Na F 34 mg/kg)fluoride exposure groups, low-dose(Na F 2.1 mg/kg+Ca CO325 mg/kg), middle-dose(Na F 8.5 mg/kg+Ca CO350 mg/kg), highdose(Na F 34 mg/kg +Ca CO3100 mg/kg) fluoride-calcium combined exposure groups,10 in each group,half male and half female. The exposure by intragastric administration lasted 90 days. The concentrations of 24 h urinary fluoride were examined every 15 days. After the last dosing,the levels of bone fluoride were measured. Results Compared with the control group,the contents of bone fluoride in low-, middle-, high-dose fluoride exposure groups and middle-, high-dose fluoride-calcium combined exposure groups increased significantly, the concentrations of urinary fluoride in low-, middle-, high-dose fluoride exposure groups and low-, middle-, high-dose fluoride-calcium combined exposure groups increased significantly(P〈0.05).The contents of bone fluoride and urinary fluoride were both increased with the increase of fluoride exposure dosages.Compared with the same dose of fluoride exposure groups,the levels of bone fluoride in fluoride-calcium combined exposure groups decreased significantly(P〈0.05); The calcium antagonism effects on urinary fluoride showed variability in different gender, fluoride-calcium combined exposure doses,exposure time. Conclusion The accumulation of fluoride in rats may be antagonized by appropriate dose of calcium.
