珍宝丸对大鼠脊髓损伤后RhoA的调节作用

摘　　要：目的在大鼠脊髓不完全损伤后,观察蒙药珍宝丸对神经的再生修复作用及其对RhoA表达的调节作用。方法利用Allen's改良方法建立脊髓不完全损伤的动物模型,把48只成年Wistar大鼠随机分为损伤组(A组,n=24)和损伤后珍宝丸治疗组(B组,n=24),B组在损伤脊髓后以珍宝丸灌胃治疗。A、B组按损伤后3、7、14 d分为3个时间点,每个时间点8只。分别在术后3、7、14 d对A、B组大鼠后肢运动功能进行BBB评分,再在各时间点处死动物,取受损脊髓行H.E染色及免疫组化染色观察形态学变化和RhoA在脊髓组织中的分布特点,同时对各组相应时间点行阳性细胞计数,并作比较。结果在7 d和14 d这两个时间点,B组的BBB评分明显高于A组,差异有统计学意义(P<0.05)。B组大鼠受损脊髓的病理形态改善。B组的RhoA蛋白表达量显著低于A组,差异有显著性(P<0.05)。结论大鼠急性脊髓损伤后RhoA表达显著升高,早期应用珍宝丸对RhoA的表达具有一定的抑制作用。Objective To investigate the effects of mongolian medicine Zhenbao pills on expression of RhoA in rats with acute incomplete spinal cord injury .Method Rat model of spinal cord injury was established by modified Allen’s method .Forty -eight adult Wistar rats were divided into the spinal cord injury group （group A ,n=24） and group B（ n=24 ） treated with Zhenbao pills at random after spinal cord injury .The rats in group B was treated with Zhenbao pills （1 g/kg） once a day by gavage .At 3 d ,7 d and 1 4 d after operation , motor functions of the rat hind limbs were determined by the BBB （Basso Beattie and Bresnahan） ,and the rats in each group were killed at 3 d ,7 d and 1 4 d post operation for histopathology examination .Morphologic changes of spinal neurons were evaluated by HE staining and immunohistochemistry ,respectively .Results It was ob-served that at 7 d and 1 4 d after operation ,the BBC score of group B was significantly higher （ P〈0 .0 5 ） than that of group A .However ,the expression levels of RhoA in group A were significantly higher （ P〈0 .0 5 ） than that in group B .Conclusion The expression of RhoA was markedly increased in rats with acute spinal cord in-jury ,and early use of Zhenbao pills may inhibit the expression of RhoA .
