ABO血型抗体效价检测用标准物质研制与初步应用

摘　　要：目的研制ABO血型抗体效价检测的标准物质,作为血型抗体效价检测的室内质控和检测结果溯源的标准。方法将标准单克隆血型抗体分别使用生理盐水、低离子液和AB型血浆进行稀释,并观察反复冻融对抗体效价的影响;将合适稀释度的血型抗体冻干制备成标准物质,并对标准物质的均一性、稳定性进行评定。结果生理盐水和低离子液稀释的标准物质冻融后抗体效价变化较大,AB型血浆稀释的标准物质冻融后效价变化不大;标准物质均一性评价的方差分析结果显示P>0.05,表明标准物质在瓶间是均匀的;35d内各参数的稳定性评价趋势分析结果均显示P>0.05,表明该期限内标准物质是稳定的。结论使用AB型血浆稀释制备标准物质较好,制备的标准物质均匀性和稳定性良好。Objective To establish the standard for the detection of blood group antibodies for the internal quality control and the traceability of the test results.Methods The standard was eatablished with dilution of monoclonal antibodies against blood group antigens by normal saline,low ionic liquid and AB type plasma,and the change in titer effected by repeated freezing and thawing,and the homogeneity and the stability were detected.Results The repeated freezing and thawing caused an obvious change in titer of the standard materials diluted by normal saline and low ionic liquid,but no change was noted in the titer of AB group plasma.The standard material was homogeineous and stable when tested within 35days（P〉0.05）.Conclusion The standard diluted by AB type plasma meets the needs of the homogeneity and stability of the certified reference materials.
