肠益煎对裸鼠结肠癌移植瘤抑瘤作用的实验研究

摘　　要：目的探究肠益煎对移植性结肠癌裸鼠肿瘤的抑制作用。方法将接种人结肠癌LOVO细胞悬液于裸鼠皮下成功制备的裸鼠移植性结肠癌模型60只随机分为模型组(A组)、肠益煎组(B组)、5-氟尿嘧啶组(C组)、5-氟尿嘧啶+奥沙利铂组(D组)、肠益煎+5-氟尿嘧啶组(E组)、肠益煎+5-氟尿嘧啶+奥沙利铂组(F组),每组10只,模拟临床给药。A组给予生理盐水20 m L/(kg·d)灌胃;其他组中肠益煎采用水煎剂12.5 g/(kg·d)灌胃,均每天1次,共14 d;5-氟尿嘧啶及奥沙利铂采用腹腔注射,隔天1次,共5次。疗程中观察并记录裸鼠体质量。疗程结束后处死裸鼠,采集标本,记录瘤质量,计算肿瘤指数和抑瘤率。结果裸鼠去瘤后质量B、C、D、E、F组均显著高于A组(P均<0.05),D、E、F组均显著高于B、C组(P均<0.05),D、E、F组间比较差异均无统计学意义;C、D、E、F组裸鼠瘤质量、肿瘤指数明显低于A、B组(P均<0.05),D、F组明显低于C组(P均<0.05)。C、D、E、F组抑瘤率均明显高于B组(P均<0.05),D、F组明显高于C组(P均<0.05)。结论中药复方肠益煎可以稳定患瘤裸鼠体质量,有一定的抑制肿瘤瘤体生长作用,联合化疗药物5-氟尿嘧啶、奥沙利铂抑瘤效果更佳。Objective It is to investigate the anti rumor effect of Changyi Decoction on nude mice with transplanted colorectal cancer（ CRC）. Methods 60 nude mice with inoculation by transplant Colorectal Carcinoma LOVO cell suspension under their skin were randomly divided into 6 groups as model control group（ Group A）,Changyi Decoction group（ Group B）,5-Fluorouracil group（ Group C）,5- Fluorouracil ＋ Oxaliplatin group（ Group D）,Changyi Decoction ＋ 5- Fluorouracil group（ Group E） and Changyi Decoction ＋ 5- Fluorouracil ＋ Oxaliplatin group（ Group F）,10 mice in each group. Simulating the clinic,the mice were treated with normal saline（ NS） in Group A,Changyi Decoction in Group B,5- Fluorouracil（ 5- Fu）in Group C,5- Fluorouracil and Oxaliplatin（ L- OHP） in Group D,Changyi Decoction and 5- Fu in Group E,Changyi Decoction,5- Fu and L- OHP in Group F. NS and Changyi Decoction were given by gavage once a day for 14 days with NS 20 m L /（ kg·d） and Changyi Decoction 12. 5 g /（ kg·d）. 5- Fu and L- OHP were given every other day by intraperitoneal injection for 5 times. In the course,mice weights were recorded. After treatment,the tumor weight was measured and tumor growth and inhibition rate were calculated. Results Mice weight after tumor removing of Group B,C,D,E and F were all markedly higher than that of Group A（ all P 〈0. 05）,and which in Group D,E,F were higher markedly than those in Group B and C（ all P 〈0. 05）,there was no significant difference among Group D,E and F（ P 〉0. 05）. The tumor weight and tumor growth rate of Group C,D,E and F were lower markedly than those of Group A and B（ all P 〈0. 05）,and which of Group D and F were higher markedly than that of Group C（ P 〈0. 05）.
