百令胶囊对肾移植大鼠肾功能及排斥反应的影响

摘　　要：目的观察百令胶囊对左肾移植大鼠肾功能及排斥反应的影响。方法将80只大鼠随机分为假手术组、模型组、环孢菌素A组、百令胶囊组,每组20只。除假手术组外,其余3组均进行同种异体左肾移植术。术后百令胶囊组给予百令胶囊20 mg/kg灌胃并腹腔注射环孢菌素A 2.0 mg/(kg·d),环孢菌素A组注射环孢菌素A 2.0 mg/(kg·d),假手术组及模型组仅灌等体积生理盐水,1次/d,均连续灌胃3周。测定治疗前后4组大鼠血清胱抑素C(Cys C)、尿素氮(BUN)、肌酐(Cr)、丙二醛(MDA)、乳酸脱氢酶(LDH)、内源性超氧化物歧化酶(SOD)、免疫球蛋白G(Ig G)、全血T淋巴细胞、单核细胞数量,并统计大鼠存活时间及存活率。结果百令胶囊组血清Cys C、BUN、Cr、MDA、LDH水平及T淋巴细胞、单核细胞数量均明显降低,血清SOD、Ig G水平升高,大鼠存活时间及存活率明显提高,与模型组及环孢菌素A组比较差异均有统计学意义(P均<0.05)。结论百令胶囊可明显抑制脂质过氧化反应,提高SOD活性,清除肾移植术后MDA,提高免疫功能,可增强环孢菌素A抗排异反应药效,促进肾功能恢复,提高大鼠存活时间及存活率,对移植肾有一定的保护作用。Objective It is to observe the influence of Bailing Capsule on renal function and rejection in rat with renal transplantation. Methods 80 rats were randomly divided into sham operation group,model group,Cyclosporin A group and Bailing Capsule group,20 cases in each. The cases in model group,Cyclosporin A group and Bailing Capsule group were performed orthotopic left kidney transplantation. Then the rats in Bailing Capsule group were treated with Bailing Capsule 20 mg /kg gavage and intraperitoneal injection of Cyclosporin A 2. 0 mg /（ kg. d） for 3- week,and with injection of Cyclosporin only in Cyclosporin A model group and saline irrigation only in sham operation group. The levels of cystatin C（ Cys C）,blood urea nitrogen（ BUN）,serum creatinine（ Cr）; serum malondialdehyde（ MDA）,lactate dehydrogenase（ LDH） and serum superoxide dismutase（ SOD）,Immunoglobulin G（ Ig G）,whole blood T lymphocyte,monocyte number were detected,and the rat survival time and survival rate were compared between two groups. Results In Bailing Capsule group,the levels of serum Cys C,BUN,Cr,MDA,LDH and the number of T lymphocyte,monocyte were decreased obviously,and the levels of serum SOD,1g G were elevated markdely,the rat survival time and survival rate were increased significantly. There were significant differences to compared with the model group and Cyclosporin A group（ all P 〈0. 05）. Conclusion Bailing Capsule can significantly inhibit lipid peroxidation,increase the activity of SOD,scavenge of MDA after renal transplantation,and enhance immunity and anti rejection drug efficacy of Cyclosporin A,reduce renal transplantation postoperative renal ischemia / reperfusion on renal function damage,promoting the recovery of renal function,improve the rat survival time and survival rate,has a protective effect on the transplanted kidney
