摘  要:目的:采用H9c2心肌细胞系,研究益心解毒方含药血清抑制血管紧张素Ⅱ(AngⅡ)诱导的心肌细胞Nox2、Nox4型NADPH氧化酶表达的影响。方法:取对数生长期的H9c2心肌细胞,同步化后进行实验,实验分为正常组、模型组、益心解毒方组、二亚苯基碘(DPI)对照组。采用乙酰乙酸双氯荧光素(DCFH-DA)法测定H9c2心肌细胞内活性氧(ROS)水平;RT-PCR法和Western-blot法测定Nox2亚基和Nox4亚基的表达量。结果:研究发现,与模型组比较,益心解毒方含药血清能抑制ROS和Nox2、Nox4亚基表达的增高,其中益心解毒方低剂量组作用最为显著(P<0.01,P<0.05)。结论:结合课题组前期的动物实验研究表明,益心解毒方在改善心功能、减缓心室重构等方面药效显著,而抑制NADPH氧化酶活性,降低心肌ROS水平可能是其发挥药效的重要机制。Objective: To study the effects of Yixin Jiedu Formula on the expression of Nox2 and Nox4 type NADPH oxidase in H9c2 cardiac muscle cells induced by angiotensinⅡ(AngⅡ).Methods: H9c2 cardiac muscle cells in logarithmic phase synchronized were divided into normal group,model group,Yixin Jiedu Formula group and diphenylene iodine(DPI) control group.The level of ROS in H9c2 cardiac muscle cells had been detected by DCFH-DA method.RT-PCR and Westernblotting method were used to measure the expressions of Nox2 and Nox4 subunit.Results: The level of ROS,the expressions of Nox2 and Nox4 type NADPH oxidase in H9c2 cardiac muscle cells induced by AngⅡ were all significantly increased,which were significantly inhibited by Yixin Jiedu formula drug serum,especially the low-dose Yixin Jiedu Formula drug serum(P〈0.01,P〈0.05).Conclusion: In combination with previous animal experiments,it shows that Yixin Jiedu Formula could significantly improve heart function and slow the ventricular remodeling,the mechanism of which may be related to inhibiting the activity of NADPH oxidase and reducing the level of ROS in cardiac muscle cells.
