仁术健脾理气方对功能性消化不良大鼠胃平滑肌收缩反应及GHSR蛋白表达的影响

摘　　要：目的:研究仁术健脾理气方对功能性消化不良(FD)大鼠的治疗作用机制。方法:雄性10日龄SD幼鼠30只,随机分为正常组(10只)和模型组(20只)。采用0.1%碘乙酰胺(IA)灌胃诱导大鼠FD模型。大鼠8周龄时进行模型评定,将造模成功的20只大鼠随机分为模型组(10只)和中药组(10只)。正常组与模型组予以0.9%氯化钠溶液灌胃,中药组给予仁术健脾理气方灌胃。采用Power lab生物信号采集系统记录离体大鼠胃体纵行平滑肌的收缩活动,采用蛋白免疫印记法(Western Blot)和免疫组织化学(IHC)技术检测生长激素促释放激素受体(GHSR)在FD大鼠胃体的表达。结果:离体胃纵行平滑肌条收缩记录结果显示中药组大鼠胃肌条的收缩平均频率和振幅变化率显著高于模型组(P<0.05)。Western Blot和IHC结果均显示中药组大鼠胃体GHSR蛋白水平显著高于模型组(P<0.01)。结论:仁术健脾理气方对IA诱导的FD大鼠具有促进胃动力作用,其机制可能与上调GHSR蛋白表达有关。Objective： To study on the effect of Renzhu Jianpi Liqi Formula on rat with functional dyspepsia（FD） and explore its mechanism.Methods： Thirty ten-day-old male rats were randomly divided into control group（n=10） and model group（n=20）.Rat FD model was established with 0.1% of iodoacetamide（IA） by gavage.With evaluation of model rats at the age of 8 weeks,the successful model of 20 rats were randomly divided into model group（n=10） and Chinese herbal medicine（CHM） group（n=10）.The control group and model group were gavaged with 0.9% of sodium chloride solution,while CHM group were gavaged with Renzhu Jianpi Liqi Formula.Power lab biological signal was used to systematically record the contractile activity in isolated rats gastric body longitudinal muscle.Western blot（WB） and immunohistochemistry（IHC） technique were used to detecte the protein expression of GHSR in FD rats.Results： Isolated gastric smooth muscle contraction records showed that contraction frequency and amplitude change of rats gastric muscle strips in CHM group were both significantly higher than those in model group（P〈0.05）.WB and IHC results showed that GHSR protein levels in CHM group were significantly higher than those in model group（P〈0.01）.Conclusion： Renzhu Jianpi Liqi Formula could promote the gastric motility in FD rats induced by IA,the mechanism of which might be related to the up-regulation of the GHSR protein level.
