摘 要:目的:观察滋肾调冲法代表方功血宁Ⅱ号方对骨髓内皮祖细胞(EPCs)促进黄体血管新生的影响,探讨该法健全黄体功能的作用机制。方法:建立大鼠骨髓移植假孕模型,随机分为移植对照组、功血宁Ⅱ号组和乌鸡白凤丸组,另取未骨髓移植大鼠为假孕对照组。在黄体早中晚期,各组处理相应数目动物,摘取卵巢。采用PCR法检测黄体中供体雄鼠来源的Sry基因的表达;原位杂交法检测Sry基因阳性细胞;原位杂交合免疫组化双标染色,观察EPCs向血管内皮细胞分化情况;免疫组化法检测黄体微血管密度(MVD)。结果:骨髓移植各组的黄体中均检测到Sry基因和双标记阳性细胞,假孕对照组未见;Sry基因的表达量均显著高于假孕对照组(P<0.01);功血宁Ⅱ号组显著升高(P<0.05)。黄体MVD各组早期较少,中期显著增多,晚期明显减少(P<0.01);功血宁Ⅱ号组黄体中期MVD显著增多(P<0.01)。结论:滋肾调冲法能够将骨髓EPCs归巢于黄体,并分化为血管内皮细胞,使黄体MVD增加,促进黄体的血管新生,从而达到健全黄体功能的作用。Objective: Observation of the effects of Gongxuening Ⅱ,the representative prescription of Zishen Tiaochong method on bone marrow endothelial progenitor cells(EPCs) to promote corpus luteum angiogenesis,to explore its mechanism in improving luteal function.Method: The pseudopregnant rat model after bone marrow transplantation was established,the rats were randomly divided into transplantation control group,Gongxuening Ⅱ group and Wuji Baifeng pills group,meanwhile the pseudopregnant rat model as the pseudopregnant control group.In the early,middle and late period of corpus luteum,all rats were sacrificed to remove the ovary.The mRNA expression of Sry in corpus luteum was detected with PCR technology.The positive cells of Sry gene were detected by in situ hybridization method.The differentiation of EPCs to vascular endothelial cell was observed with in situ hybridization immunohistochemistry double staining method.The corpus luteum micrangium density(MVD) was detected with immunohistochemistry method.Results: The positive cells of Sry in corpus luteum gene were detected in each group except for pseudopregnant control group.The mRNA expressions of Sry in each group were all higher than those in the pseudopregnant control group(P〈0.01),with Gongxuening Ⅱ group the highest.The MVD was lower in the early period of corpus luteum,and higher in the middle period,significantly decreased in the late period(P〈0.01).In the late period,corpus luteum MVD in Gongxuening Ⅱ Group was higher than that in others(P〈0.01).Conclusion: Zishen Tiaochong method can home to the bone marrow endothelial progenitor cells to corpus luteum,and differentiate these cells to endothelial cells,which can increase the micrangium density of corpus luteum,and promote corpus luteum angiogenesis,to improve corpus luteum function.
