金磁纳米粒子功能化探针的制备及在毒素检测中的应用

摘  要:利用金磁纳米粒子(gold-magnetic nanoparticle)兼有纳米金颗粒与磁性纳米粒子特性的优势,以相思子毒素(abrin) 为目标物,将蛋白A(SPA) 包被金磁纳米粒子并偶联多抗作为功能化捕获探针,成功地建立了检测毒素的双抗体夹心磁免疫分析法,其线性范围为0.031-125g/L,回归方程为Y=0.571X+0.968 (R=0.994, n=13, P约0.0001),检出限为0.031g/L.该法适用于水样、土样、食品及血液等模拟样品中微量abrin分析,具有较好的重现性与特异性,与传统的磁性纳米粒子功能化探针相比,经蛋白A定向固定抗体制备的金磁纳米粒子功能化探针具有更高的结合活性,从而提高了检测灵敏度.Taken abrin as the target, "gold-magnetic nanoparticle-staphylococcus protein A (SPA)-polyclonal antibody" complex was used to functionalize capture probe. Double antibody sandwichmagnetic immunoassay to detect toxin was established successfully.A linear relationship betweenabsorbance value and the concentration of abrin in the range of 0.031~125滋g/L was obtained. Thelinear regression equation was Y=0.571X+0.968 (R=0.994, n=13, P约0.0001) with a detection limit of0.031滋g∕L. This method had high specificity and acceptable reproducibility for detection of analogsample such as water, soil, food and blood. In comparison with the traditional functionalized magneticnanoparticle probe, functionalized gold-magnetic nanoparticle probe possessed higher binding activity ,thus improved the sensitivity of detection.
