摘 要:目的探讨心房钠尿肽(ANP)信号通路在小鼠支气管哮喘(BA)发病中的作用及其机制。方法 40只雌性BALB/C小鼠随机分为正常对照组、BA组、ANP组和ANP+A71915组,每组10只;建立BA小鼠模型和ANP在体干预模型,观察各组小鼠支气管炎症改变情况和支气管黏液分泌情况,采用免疫组织化学技术检测肺组织T-bet和GATA-3蛋白的表达。结果 BA组小鼠支气管黏膜充血水肿及支气管周围炎性细胞浸润较正常对照组显著(P<0.01),ANP组小鼠支气管黏膜充血水肿及支气管周围炎性细胞浸润较BA组更严重(P<0.01),ANP+A71915组小鼠支气管黏膜充血水肿及支气管周围炎性细胞浸润较ANP组明显减轻(P<0.01)。BA组小鼠肺组织中GATA-3蛋白阳性表达显著高于正常对照组(P<0.01),ANP组小鼠肺组织中GATA-3蛋白阳性表达显著高于BA组(P<0.01),ANP+A71915组小鼠肺组织中GATA-3蛋白阳性表达显著低于ANP组(P<0.01)。BA组小鼠肺组织中T-bet蛋白表达显著低于正常对照组(P<0.01),ANP组小鼠肺组织中T-bet蛋白表达明显低于BA组(P<0.01),ANP+A71915组小鼠肺组织中T-bet蛋白表达明显高于ANP组(P<0.01)。结论 ANP信号可能通过调控T-bet和GATA-3蛋白的表达参与BA小鼠免疫功能失衡的发病过程。Objective To discuss the effect and its mechanism of atrial natriuretic peptide( ANP) signaling pathway in the pathogenesis of bronchial asthma( BA) in mice. Methods Forty female BALB / C mice were divided into normal control group,BA group,ANP group and ANP + A71915 group,ten mice in each group. The BA mice models and ANP intervention models were established. The inflammation and mucus secretion of bronchus of mice were observed. The expression of T-bet and GATA-3 protein in lung tissue was detected by immunohistochemical method. Results The bronchial mucosal congestion and edema and peribronchial inflammatory cell infiltrate in BA group were significantly more than those in normal control group(P 0. 01). The bronchial mucosal congestion and edema and peribronchial inflammatory cell infiltrate in ANP group were significantly more seriously than those in BA group( P 0. 01). The bronchial mucosal congestion and edema and peribronchial inflammatory cell infiltrate in ANP + A71915 group were significantly less than those in ANP group( P 0. 01). The positive expression of GATA-3 in lung tissue of mice in BA group was significantly higher than that in normal control group( P 0. 01). The positive expression of GATA-3 in lung tissue of mice in ANP group was significantly higher than that in BA group(P 0. 01). The positive expression of GATA-3 in lung tissue of mice in ANP + A71915 group was significantly lower than that in ANP group( P 0. 01). The expression of T-Bet protein in lung tissue of mice in BA group was significantly lower than that in normal control group( P 0. 01). The expression of T-Bet protein in lung tissue of mice in ANP group was significantly lower than that in BA group( P 0. 01). The expression of T-Bet protein in lung tissue of mice in ANP + A71915 group was significantly higher than that in ANP group( P 0. 01). Conclusion ANP signal may be involved in the pathogenesis of immune imbalance of mice with BA by regulating the expression of T-bet and GA
