摘 要:目的:制备水杨酸甲酯立方液晶并对其进行表征,同时建立水杨酸甲酯含量测定方法。方法:以植烷三醇为基质材料,采用自乳化方法制备水杨酸甲酯立方液晶;用偏光显微镜及小角衍射(SAXS)对立方液晶进行表征;采用HPLC测定立方液晶中水杨酸甲酯的含量。结果:立方液晶在偏光显微镜下视野黑暗;小角衍射显示在1.46,1.79,2.05,2.50处出现信号峰,其比值q1∶q2∶q3∶q4=21/2∶31/2∶41/2∶61/2,即为立方相液晶,其内部结构为Pn3m(Q224)双菱形晶格。水杨酸甲酯在0.99~198.12μg·mL-1呈良好线性关系(r=0.999),平均回收率为99.89%,RSD为0.58%。结论:偏光显微镜及小角X射线衍射适于立方液晶的表征,高效液相测定方法简便、可行。Objective: To prepare and characterize cubic liquid crystalline as drug delivery systems for methylsalicylate,and establish a method for determination of content of methylsalicylate. Methods: Cubic liquid crystal was prepared by self-emulsification of phytantriol in water; the liquid crystaline structure was confirmed by polarizing microscopy and small-angle X-ray diffraction; an HPLC method was developed to determine the content of methylsalicylate. Results: Cubic liquid crystalline showed dark background under polarizing microscopy. The values of the scattering vector q corresponding to the scattering peaks were 1. 46,1. 79,2. 05,and 2. 50,the ratio of q1∶ q2∶ q3∶ q4 was 21/2∶31/2∶41/2∶61/2,which was characteristic of cubic periodicity indicative of a diamond( Q224). The linear range of the calibration curve for methylsalicylate was 0. 99 ~ 198. 12 μg·m L- 1( r = 0. 999),the average recovery of methylsalicylate was 99. 89% with RSD of 0. 58%. Conclusion: Polarizing microscopy and small-angle X-ray diffraction can be used to characterize cubic liquid crystalline. This determination method is convenient,accurate,and reproducible,which provides a determination method of methylsalicylate content.
