苏木乙酸乙酯提取物对慢性病毒性心肌炎模型小鼠T细胞亚群的影响

摘　　要：目的:观察苏木乙酸乙酯提取物对慢性病毒性心肌炎小鼠T细胞亚群的影响,从而探讨其作用机制,为治疗慢性病毒性心肌炎提供客观的理论依据。方法:采用多次腹腔注射CVB3病毒的方式,复制重复感染慢性病毒性心肌炎动物模型。将145只Balb/C小鼠随机选取10只做为空白组,选取135只小鼠为模型组、于第0、15、30天腹腔注射CVB3病毒,剂量分别为0.1ml、0.15ml、0.2ml,第45天后将存活小鼠选取75只进行分组,随机分为模型组、环孢素组、苏木组,每组各25只分组后进行给药,连续给药20天。采用天狼星红染色观察其心肌病理形态及超微结构,采用流式细胞仪检测外周血中CD+4T、CD+8T细胞含量,并计算CD+4/CD+8T的比值。结果:病理结果显示,苏木乙酸乙酯提取物能改善CVB3感染小鼠的心肌病理损伤。结论:苏木乙酸乙酯提取物能够降低慢性病毒性心肌炎小鼠CD+4、CD+4/CD+8T水平,升高CD+8T水平,这可能是其治疗病毒性心肌炎的机制之一;苏木乙酸乙酯提取物能改善病毒感染心肌组织的病理损伤,对心肌组织具有一定的保护作用。Objective： To study the mechanism of the impact of sppan acetic ether extract on T cell subgroup of mice with chronic viral myocarditis,and to provide an objectively theoretical basis for the treatment of chronic viral myocarditis.Methods： By giving intraperitoneal injection of CVB3 virus many times,duplicate the repeatedly infected animal models with chronic viral myocarditis.Select 10 Balb / C mice as a blank group from 145 mice randomly; other 135 mice were chosen as a model group,with intraperitoneal injection of CVB3 virus on 0,15 th,30th days,the doses on different day were 0.1ml,0.15 ml and 0.2ml.Select 75 survivors and divide them into a model group,a cyclosporine group,and the Sppan group randomly.Each group had 25 mice,and give them injection continuously for 20 days.Observe the pathological and ultrastructural changes of cardiac muscle stained with Sirius-Red.Detect the content of CD＋4T and CD＋8T cells in Peripheral blood with flow cytometry.Calculate the ratio of CD＋4/ CD＋8T.Results： Pathological results showed that Sppan acetic ether extract could improve the myocardial pathological damage of the mice that were infected by CVB3.Conclusion：Sppan acetic ether extract can reduce the levels of CD＋4,CD＋4/ CD＋8T in chronic viral myocarditis mice and improve the levels of CD＋8T.This may be one of the mechanism for treating viral myocarditis.Sppan acetic ether extract can improve pathological injury of infective myocardial tissue,which has certain protective effect on myocardial tissue.
