摘 要:目的:观察华蟾素注射液对H22肝癌小鼠肿瘤组织Smac(半胱氨酸)和Caspase-9(天冬氨酸蛋白酶)基因(m RNA)及蛋白表达的影响。方法:40只ICR小鼠采用皮下接种H22肝癌细胞株的方法复制小鼠肝癌模型;造模成功后,将小鼠随机分为4组:模型组,华蟾素注射液低、高剂量组,顺铂(DDP)组;观察各组小鼠肿瘤生长情况变化,采用荧光定量RT-PCR方法和Western Blot方法检测各组H22小鼠肿瘤组织中Smac、Caspase-9基因及蛋白表达。结果:华蟾素低剂量组不能有效抑制肿瘤的生长,而华蟾素高剂量组、DDP组的抑瘤率分别为43.13%、47.06%,具有明显的抑制肿瘤生长作用(P<0.01)。与模型组比较,华蟾素低剂量组小鼠肝癌组织中的Smac、Caspase-9表达无明显差异,而华蟾素高剂量组、DDP组能明显促进肝癌组织中的Smac、Caspase-9的m RNA及蛋白表达(P<0.01)。结论:华蟾素注射液具有抑制肝癌细胞生长的作用,上调肿瘤组织Smac、Caspase-9基因及蛋白的表达,促进肝癌细胞发生凋亡是其部分作用机制。Objective: To investigate the effects of Cinobufacini Injection on Smac、Caspase- 9 genene( m RNA) and proteinin expressions in tumor tissues of mice bearing Hepatoma H22. Methods: Forty ICR mice were randomly divided into 4 groups: the model group,low,high dosage groups of Cinobufacini Injection,Cisplatin group,after the mice model bearing Hepatoma H22 were established by subcutaneous inoculating tumor cells,tumor growth in mice was observed,the m RNA and protein expressions of Smac,Caspase- 9 in tumour tissues were detected by quantitativereal- time reverse- transcription polymerase chain reaction( RT- PCR) and Western Blot. Results: The low dosage of Cinobufacini Injection could not inhibit the growth of tumor,the cancer inhibition ratio of high dosage Cinobufacini Injection and Cisplatin group was 43. 13% 、47. 06% respectively,itshowed obvious inhibitory effect on tumor growth( P〈0. 01). Compared with the model group,the expression levels of Smac,Caspase- 9 m RNA and protein in low dosage Cinobufacini Injection group has not obvious difference,while the high dosage Cinobufacini Injection and Cisplatingroup could promote the expressions of Smac,Caspase- 9 m RNA and protein( P〈0. 01) in tumor tissues of mice. Conclusion: Compbifacoinit Injection can inhibi the liver cancer growth by up- regulating the expressions Smac and Caspase- 9,to promote tumor cell apoptosis,and achieve anti- tumor effect.
