摘  要:目的:多指标综合优选芪归银方的水提醇沉工艺。方法:以黄芪甲苷、绿原酸和干膏得率的综合评分为指标,采用正交试验筛选出最佳水提取工艺;以黄芪甲苷、绿原酸和总多糖为指标,单因素考察醇沉浓度和药液浓缩相对程度对醇沉工艺的影响。结果:最佳水提醇沉工艺为12倍量加水量、提取3次,每次1 h,合并提取液,浓缩至1.0 g/m L生药,60%醇沉。结论:优选出来的水提醇沉工艺稳定可行,为工艺的规范化和进一步研究提供了科学依据。Objective: The technique of water extraction and alcohol precipitation of Qiguiyin formula was optimized.Method: The amount of water,extraction times and time of extraction process were investigated in the orthogonal text with the contents of astragaloside,chlorogenic acid and extract yield as index. With the contents of astragaloside,chlorogenic acid and polysaccharide as index,the influencing factors including the concentration of alcohol precipitation and relative density of concentrate on alcohol precipitation were considered in alcohol precipitation technique. Result: Optimum water extraction and alcohol precipitation technology was as following: extracted 3 times with 12 folds the amount of water,1 h for each time. The extracts were combined and concentrated to 1. 0 g·ml^-1 with the concentration of alcohol precipitation60%. Conclusion: Optimized water extraction and alcohol precipitation technology was reasonable and feasible and it could provide a reference for standardized production and further development of Qiguiyin Formula.
