丹参酮ⅡA对拉应力下人足跟成纤维细胞增殖及分泌TGF-β1、IL-6的影响

摘　　要：目的:观察丹参酮ⅡA对拉应力刺激下的人足跟外侧皮肤成纤维细胞增殖抑制的影响,为预防跟骨骨折术后切口边缘皮肤坏死、切口不愈合提供一定的理论依据。方法:取人足跟外侧皮肤成纤维细胞培养及传代后,分拉应力组、低剂量组、中剂量组、高剂量组、对照组进行拉应力加载试验,检测培养液中TGF-β1、IL-6含量,使用透射电镜、光镜观察线粒体、细胞核的变化。结果:在10%的拉应力下,成纤维细胞的线粒体减少,TGF-β1和IL-6的分泌受到抑制,细胞增殖受到抑制;丹参酮ⅡA能使成纤维细胞的线粒体增加,TGF-β1和IL-6的分泌增加,促进成纤维细胞的增殖。结论:人足跟外侧皮肤长时间处于拉应力状态是跟骨骨折手术后切口边缘皮肤坏死、切口不愈合的重要原因。丹参酮ⅡA有利于跟骨骨折手术后切口的愈合。Objective： To observe the effect of tanshinone ⅡA on the lateral heel skin fibroblast proliferation under tensile stress,providing a theoretical basis for the wound edge skin necrosis and nonunion after calcaneal fracture surgery.Methods： Fibroblast cells were taken from the 40 years old males＇ lateral heel skin,and then cultured and subcultured in vitro. After that,they were divided into five groups： tensile stress group,low dose group,middle dose group,high dose group and control group and tested by tensile stress testing. We measured the levels of TGF-β1 and IL-6 in nutrient fluid. Using transmission electron microscope,we studied mitochondria and nucleus. Results： Under 10% of the tensile stress,mitochondria decreased and the levels of TGF-β1 and IL-6 in nutrient fluid decreased and cell proliferation was inhibited. Under tanshinone Ⅱ A,mitochondria increased and the levels of TGF-β1 and IL-6 in nutrient fluid increased and cell proliferation was promoted. Conclusions： The human lateral heel skin in a long-time tensile stress state is an important cause of wound edge skin necrosis and nonunion after calcaneus fracture surgery. Tanshinone is conducive to the healing of incision after operation of fracture of calcaneus.
