摘  要:目的:通过研究一次急性运动后肝跨膜丝氨酸蛋白酶6(transmembrane serine proteases 6,TMPRSS6)、抗菌多肽(Hepcidin)、肝膜铁转运蛋白1(Ferroportin 1,FPN1)和机体铁状态的时相性变化,阐明TMPRSS6-Hepcidin-FPN1调控通路在急性运动中对机体铁代谢的调控作用。方法:30只雄性SD大鼠分为对照组(control group,CG组)6只、运动组24只(30 m/min,运动至力竭)。运动组分别于运动后0h、3h、6h、24h取材,分别称为E0h组、E3h组、E6h组、E24h组,每组6只。试剂盒检测血清铁和总铁结合力,Western blot法检测FPN1的蛋白表达,RT-PCR法测定大鼠肝脏TMPRSS6、铁调素调节蛋白(hemojuvelin,HJV)、Hepcidin mRNA。结果:(1)血清铁:E3h组与CG组相比显著降低(P<0.01),E6h组显著高于其他各运动组(P<0.01)。(2)FPN1蛋白:E3h组与CG组相比显著降低(P<0.01),E24h组与其他各运动组相比显著升高(P<0.01)。(3)TMPRSS6 mRNA:E3h组显著低于其他各组(P<0.01),E24h组显著高于E3h和E6h组(P<0.01)。HJV mRNA:E3h组显著高于其它各组(P<0.01),E24h组显著低于E0h组(P<0.05)。Hepcidin mRNA:E3h组和E6h组显著高于其它各组(P<0.01)。结论:急性运动后血清铁和总铁结合力出现明显的时相性变化,而肝的铁代谢调控基因TMPRSS6、HJV、Hepcidin及FPN1也具有相应的时相性变化,说明TMPRSS6-Hepcidin-FPN1通路对急性力竭运动后机体铁状态的调控发挥了重要作用,其影响机制还需深入探讨。Objective The time phase of transmembrane serine proteases 6(TMPRSS6),hepcidin,ferroportin 1(FPN1)and iron status of rats after an acute exhausted exercise were observed.Methods 30 male SD rats were randomly divided into control group(C),and exercise groups(groups E0 h,E3h,E6 h,and E24 h,corresponding to their sampling time of 0 hour,3 hours,6 hours,and 24 hours,respectively after exercise).The serum iron(SI)and total iron binding capacity(TIBC)were observed by Sigma kit,the TMPRSS6,hemojuvelin(HJV),and hepcidin mRNAs by RT-PCR,and expression of FPN1 by Western blot.Results(1)The SI content of group E3h(33.82 ± 12.32μmol/L)decreased significantly compared with that of group C(87.83 ± 6.31μmol/L)(P 0.01).The SI content of group E6h(93.75 ±11.04μmol/L)was significantly higher than the groups EOh(55.28 ± 14.94μmol/L)and E24h(63.49 ±8.75μmol/L)(P 0.01).(2)Compared to group C,the expression of FPNl protein in group E3 h decreased significantly(P 0.01).The expression of FPNl protein in group E24 h was significantly higher than the other exercise groups(P 0.01).(3)The expression of TMPRSS6 mRNA in group E3 h decreased significantly comparing to the other groups(P 0.01),while increased significantly in group E24 h comparing to groups E3 h and E6h(P 0.01).The expression of HJV mRNA in group E3 h increased significantly comparing to the other exercise groups(P 0.01),and in group E24 h decreased significantly comparing to group E0h(P 0.05).The expression of hepcidin mRNA in groups E3 h and E6 h increased significantly comparing to the other groups(P 0.01).Conclusion TMPRSS6-hepcidinFPN1 pathway might play an important role in the regulation of iron metabolism after exhaustive exercise in rats and further study needs to be done in this area.
