摘  要:目的研究左、右归丸含药血清经转化生长因子(TGF)-β1/Smad4信号途径对骨髓间充质干细胞(BMSCs)成骨诱导的影响。方法以左归丸、右归丸水煎液和阳性对照药补佳乐、蒸馏水给SD大鼠灌胃后取血制备含药血清(含有10%含药血清的诱导剂),与诱导剂组共5种处理因素对BMSCs进行干预,选用Western印迹法检测碱性磷酸酶(ALP)蛋白的表达,用免疫组化SP法检测TGF-β1和Smad4蛋白的表达。结果左、右归丸与空白组和诱导剂组比可显著促进ALP蛋白的表达,增强TGF-β1和Smad4蛋白的表达(P<0.01),左归丸组与右归丸组比较可上调ALP、Smad4蛋白的表达(P<0.01)。结论左归丸与右归丸可能通过增强ALP、TGF-β1、Smad4蛋白的表达,影响TGF-β1/Smad4信号途径,从而协同诱导剂促进BMSCs的成骨诱导,并且左归丸较右归丸促进作用更明显。Objective To observe the effects of serum containing ZuoGuiWan and Youguiwan on the osteogenic induction in bone marrow mesenchymal stem cells ( BMSCs) through transforming growth factor ( TGF)-β1/Smad signaling pathway .Methods Containing se-rum of Zuoguiwan,Youguiwan,positive control drug (Estradiol Valerate,BJL)and distilled water were prepared to separately add inducer , with inducer group,simultaneously intervene the BMSCs.The expression of alkaline phosphatase (ALP) protein was determined by Western blot,the expression of TGF-β1and Smad4 protein were tested by immunohistochemical streptavidin-perosidase method.Results Compared with that of control and inducer groups ,Zuiguiwan and Youguiwan remarkablely promoted the expression of ALP protein ,upregulated the ex-pressions of TGF-β1 and Smad4 protein (P〈0.05).Compared with Youguiwan group ,Zuoguiwan group enhanced the expressions of ALP and Smad4 protein(P〈0.05).Conclusions Through ALP enhancement the expressions of TGF-β1 and Smad4 protein,Zuiguiwan and Yougui-wan could effect TGF-β1/Smad4 signaling pathways ,thus cooperate with inducer on the osteogenic induction in BMSCs .
