摘 要:目的:通过对兔建立冠心病(CHD)及再狭窄(RS)模型,探讨环孢素A(cyclosporin A,CSA)在CHD及再狭窄发生发展过程中是否具有干预作用。方法:将48只兔随机分为正常对照(N)组、N+CSA组、CHD组、CHD+CSA组、RS组及RS+CSA组,每组8只。取耳缘静脉血检测免疫球蛋白水平。并对血管标本进行染色和血管标本炎症因子MMP-1、MMP-9、VCAM-1、IL-6和TNF-αmRNA表达的测定。结果:1各实验终点时,CHD组及RS组血清IgE、IgG、IgM水平较实验前比较差异无统计学意义(P>0.05),与N组比较差异亦无统计学意义(P>0.05),予环孢素A干预后未见显著变化(P>0.05)。2CHD及RS组病变髂动脉的MMP-9、VCAM-1、TNF-α的表达较N组呈升高趋势,MMP-1、IL-6的表达较N组呈减低趋势,各组整体比较差异无统计学意义(P>0.05),予CSA干预后未见显著改变(P>0.05)。结论:在本实验中未证实环孢素A对兔CHD组及RS组具有有益的干预作用,CSA用于CHD的防治有待进一步实验研究。Objective:In this research,through establishing rabbits coronary heart disease(CHD)and restenosis(RS)model,to disscuss the intervention function of cyclosporin A(CSA)on the immunologic mechanism of CHD and RS,for providing a new way of the prevention and treatment of the atherosclerotic disease.Method:Fortyeight 48 rabbits were randomly divided into normal control group(N group)and normal breeding+cyclosporine A group(N+CSA group),coronary heart disease model group(CHD),coronary heart disease+cyclosporine A group(CHD+CSA group),restenosis model group(RS group)and restenosis+cyclosporine A(RS+CSA group).The peripheral venous blood were taken to measure the immunoglobulins.Vascular specimens were dyed,and the in inflammation factors MMP-1,MMP-9,VCAM-1,IL-6and TNF-αmRNA expression of vascular specimens were determined.Result:1Compared with the beginning of the study,there were no statistical differences of changes in IgE,IgM,IgG at the groups of CHD and RS at the ending(P〉0.05).The groups with CSA,levels of Igs had no statistical differences(P〉0.05).2Compared with groups of normal food,the expression of MMP-9,VCAM-1,TNF-αtended to increase,and the expression of MMP-1,IL-6showed a decreased trendm,however there were no statistical differences(P〉0.05).After the intervention of CSA,the expression changes of these inflammatory factors were no statistical differences(P〉0.05).Conclision:In this study,the benefit of CSA on the immunologic mechanism of CHD and RS could not be confirmed,and the mechanism of CSA on CHD need to be studied intensively.
