血脂康对腹主动脉缩窄高血压大鼠左心室肥大的影响

摘　　要：目的:本研究拟探讨血脂康对腹主动脉缩窄(AAC)高血压大鼠左心室肥大的影响。方法:构建腹主动脉缩窄的高血压大鼠模型,采用超声心动图、实时定量PCR和酶联免疫分析法(ELISA)观察血脂康对大鼠左心室肥大的影响。结果:血脂康显著降低AAC高血压大鼠心重/体重比(HW/BW)、左心室重指数(LVW/BW)、收缩期心室前壁厚度(AWTd)、舒张期心室前壁厚度(AWTs)和舒张期心室后壁厚度(PWTd);并可显著降低AAC高血压大鼠左心室中脑利尿肽(BNP)和β-心肌肌球蛋白重链(β-MHC)的mRNA水平以及炎症因子白介素6(IL-6)、C-反应蛋白(CRP)和肿瘤坏死因子α(TNF-α)的蛋白水平。结论:血脂康可显著改善腹主动脉缩窄高血压大鼠左心室肥大,其机制与促炎症因子IL-6、CRP和TNF-α的表达降低有关。Objective：To explore whether IL-6,CRP and TNF-αwere involved in XZK reversing or inhibiting left ventricular hypertrophy in AAC-induced hypertensive rats.Method：Abdominal aortic constriction（AAC）rats were used to explore the effect of XZK on left ventricular hypertrophy through echocardiography,real-time PCR and ELISA.Result：The ratio of HW/BW and LVH/BW,anterior wall thickness and posterior wall thickness during diastole were significantly increased compared with sham group,whereas XZK could reverse or inhibit the effect.The results of real-time PCR and ELISA showed that XZK could reduce the mRNA levels of BNP andβ-MHC,and the protein levels of IL-6,CRP and TNF-αin the left ventricular myocardium tissue.Conclusion：XZK could improve left ventricular hypertrophy in AAC-induced hypertensive rats,the mechanism of which is related to the reduced expression of pro-inflammatory cytokines,including IL-6,CRP and TNF-α.
