摘　　要：目的研究早期糖尿病大鼠视网膜中热休克蛋白27（Hsp27）的表达。方法实验研究。对2013年5-8月在福建省眼科研究所将84只SD大鼠用数字法随机分为两组，糖尿病视网膜病变（DR）组与对照组，建立STZ诱导的糖尿病大鼠视网膜病模型，应用免疫组织化学与逆转录PCR从转录和翻译水平观察Hsp27在糖尿病SD大鼠（1、2、4周）视网膜上表达的变化情况。结果Hsp27蛋白及mRNA在对照组视网膜中不表达或弱表达，在DR组中表达呈进行性增强，差异有统计学意义（P〈0．05），Hsp27蛋白主要表达在视网膜神经节细胞层、内外核层及内界膜等。结论Hsp27参与早期糖尿病视网膜病缺血缺氧与氧化应激的发病机制，对视网膜与视神经细胞起到内源性的保护作用。Objective To detect the expression of Hsp27 in the retina of diabetic rats, and to in vestigate the role of Hsp27 in the pathogenesis of diabetic retinopathy （DR）. Methods Eightyfour SD rats were randomly divided into two groups： control group and DR group. Diabetes was induced by streptozotocin （STZ） intraperitoneal injection in DR group. The techniques of immunohistochemi cal and reverse transcription polymerase chain reaction （RTPCR） were used to examine the expres sion of Hsp27 protein and mRNA in retina respectively at 1, 2, 4 weeks after STZ injection. Re sults The result of immunohistochemical and RTPCR showed that the expression of Hsp27 protein and mRNA in rats retina were not found in control group, but were found gradually increased in DR group （P 〈0.05）. Conclusions Enhanced expression of endogenous Hsp27 involves in the isch emia hypoxia and oxidative stress, may play a critical role in the pathogenesis of early stage DR.
