摘  要:SIRT1(silent mating type information regulation 2homolog1)是一种具有NAD+依赖性去乙酰化酶活性的转录调节因子,参与能量代谢的调节。利用实时荧光定量PCR技术,对10月龄山西肉用绵羊母本品系去势公羊内脏(肝、肾、脾、肺、心)、肌肉(背最长肌)和脂肪(大网膜、小网膜、肠系膜、腹膜后、皮下)等11种器官或组织中SIRT1基因mRNA的表达量进行检测,以探讨SIRT1基因的差异性表达规律。结果表明,绵羊SIRT1基因在所检测的器官或组织中均有表达,且差异极显著(P<0.001)。主要表现在脾脏、肝脏和肾脏中的表达显著高于心脏和肺脏中的表达;深层脂肪组织中的表达高于浅层的皮下脂肪;在背最长肌中也有较高表达。这些差异与绵羊SIRT1基因调节脂质代谢及糖异生有关。有关结果为研究SIRT1基因的功能奠定了科学依据。SIRT1 (silent mating type information regulation 2 homolog 1) is a transcription regulation factor of NAD+ dependent deacetylase activity and associates with the regulation of energy metabolism. Wethers at 10 months of age of Shanxi Meat Sheep Dam Line were used as the experimental animals. Samples from viscera (liver, kidney spleen, lung and heart), muscle (longissimus dorsi) and adipose tissues (great and small omental, mesenteric, retroperitoneal and subcutaneous) were collected after slaughtering. Quantitative real-time PCR was used to detect the abundance of SIRT1 mRNA in these organs and tissues for exploring the differential expression pattern. The results showed that o- vine SIRT1 mRNA expressed in all the studied samples and expression difference was significant (P〈0. 001). Higher expressions in spleen, liver and kidney than in heart and lung for organs, and in deep adipose depots than in subcutane- ous adipose tissues were found. Expression with medium degree in longissimus dorsi was also detected. These differ- ential expressions of SIRT1 are associated with its functions in lipid metabolism and gluconeogenesis. Results obtained in this study can provide scientific basis for researches on the function of ovine SIRT1 gene.
