消痰通腑方对结直肠癌前病变小鼠IL-6、AICD表达的影响

摘　　要：目的探讨白细胞介素6(IL-6)与活化诱导胞嘧啶核苷脱氨酶(AICD)在结直肠癌前病变表达的关系及消痰通腑方的干预作用。方法 AOM/DSS联合造模法复制小鼠结直肠癌前病变模型,随机分为空白组、模型组、中药组、西药组、中西医结合组。空白组、模型组给予等量蒸馏水;中药组给予消痰通腑方(26 g·kg-1·d-1)浓缩液0.2 ml/d;西药组每只小鼠给予柳氮磺吡啶(7.8 mg/d);中西医结合组同时给予消痰通腑方和柳氮磺吡啶。各组均连续给药12周,HE染色观察小鼠肠黏膜上皮组织形态及不典型增生,免疫组化及酶联免疫吸附测定法(ELISA)检测IL-6、AICD表达情况。结果与正常组比较,模型组IL-6和AICD免疫组化指数、血清蛋白表达升高,差异有统计学意义(P<0.01)。与模型组比较,18周中药组、西药组、中西医结合组IL-6和AICD免疫组化指数、血清蛋白表达下降,差异有统计学意义(P<0.05)。与西药组比较,18周中药组AICD的免疫组化指数表达下降,差异有统计学意义(P<0.05)。结论消痰通腑方能够降低肠道内炎症因子IL-6及AICD的表达,从而降低发生体细胞高频突变(SHM)和类别转换重组(CSR)的机率,达到防治结直肠癌前病变的作用。Objective To investigate the relationship between interleukin 6（ IL-6） and activation-induced cytidine deaminase（ AICD） in precancerous lesions in colorectal cancer,to investigate the prevention and treatment role of ＂Xiaotan Tongfu Decoction＂in the treatment of precancerous lesions in colorectal cancer.Methods Based on the classic AOM / DSS model of precancerous lesions in colorectal cancer,C57 BL /6 mice were randomly divided into control group,model group,TCM（ ＂Xiaotan Tongfu Decoction＂） group,WM（ sulfasalazine） group,integrative medicine group. Control group and model group were given the same amount of distilled water. TCM group were given medicine＂Xiaotan Tongfu Decoction＂26 g /（ kg. d）,concentrate 0. 2ml / d; WM group were given sulfasalazine7. 8 mg / pcs / day. Integrative group were given both＂Xiaotan Tongfu Decoction＂and sulfasalazine. Continuous administration of 12 w. Results Compared with the normal group,the IL-6 and AICD immunohistochemical index,serum protein expression in the model group were increased,the difference was statistically significant（ P 〈 0. 01）. Compared with the model group,18 w IL-6 and AICD immunohistochemical index,serum protein in the TCM medicine group,WM group,Integrative Medicine group were decreased,the difference was statistically significant（ P 〈 0. 05）. Compared with WM group,18 w TCM group AICD immunohistochemical expression index decreased,the difference was statistically significant（ P 〈 0. 05）. Conclusion ＂Xiaotan Tongfu Decoction＂reduce the expression of inflammatory cytokines IL-6 and AICD,thereby reducing the occurrence of somatic hypermutation（ somatic hypermutation,SHM） and class switch recombination（ class-switch recombination,CSR）,playing the role of prevention and treatment of Precancerous lesions in colorectal cancer.
