摘  要:目的研究白藜芦醇(resveratrol,Res)对脑缺血再灌注(ischemia/reperfusion,I/R)损伤保护作用的可能机制。方法 90只大鼠随机平均分为9组,假手术组(A组)、术后持续缺血组(B1组)和缺血再灌注后丙二醇溶液组(B2组)、持续缺血后Res干预组(C1~C3组)和缺血再灌注后Res干预组(D1~D3组),Res浓度分别为10~8 g/kg、10~7 g/kg、10~6 g/kg。大鼠脑缺血2 h后再灌注,之后Res治疗3天。反相高效液相层析检测海马组织谷氨酸(glutamate,Glu)含量,免疫组化方法测定谷氨酸/天冬氨酸转运体(glutamate/aspartate transporter,GLAST)及谷氨酸转运体-1(glutamate transporter-1,GLT-1)蛋白阳性的表达。结果与B1~B2组比较,C1~C3组和D1~D3组Glu含量明显降低,同时GLAST和GLT-1的表达升高。结论 Res通过提高GLAST和GLT-1的表达,降低Glu含量,起到脑缺血再灌注损伤后的神经保护作用。Objective To explore the effect of resveratrol( Res) on hippocampal of brain in ischemia / reperfusion rats. Methods The middle cerebral artery occlusion models of rats were made. Ninety male Sprague-Dawley rats were randomly divided into 9 groups( n = 10) : sham operation group( A group),continuing ischemia group( B1 group) and propylene glycol solution after reperfusion group( B2 group),Res after continuing ischemia group( C1 ~ C3groups) and Res after reperfusion group( D1 ~ D3 groups),and the dose of Res was 10 ~ 8 g /kg,10 ~ 7 g / kg,10 ~ 6 g / kg respectively. 3d Res treatment after reperfusion,reverse phase high-performance liquid chromatography( RP-HPLC) was used to examine the glutamate concentration of the ischemic brain tissue homogenate,and immunohistochemistry was used to examine the expression of glutamate / aspartate transporter( GLAST) and glutamate transporter-1( GLT-1). Results Compared with B1 ~ B2 groups,the glutamate concentration was significantly decreased( P 0. 05),and the expression of GLAST and GLT-1 was increased in C1 ~C3 groups and D1 ~ D3 groups. Conclusion The mechanism of neuroprotective effects of Res on focal cerebral ischemia / reperfusion injury may by increasing the expression of GLAST and GLT-1 in the brain tissue and inhibiting the expression of glutamate.
