摘 要:目的 :探讨紫杉醇-壳寡糖缓释膜通过抑制TGFβ/Smad信号对良性胆管瘢痕纤维化产生抵抗作用的机制及其最佳作用因素。方法:提取新西兰大耳白兔的成纤维细胞进行体外培养分组,分别加入不同浓度的紫杉醇溶液,通过MTT法测定成纤维细胞的生长活性,IFCC测定α-SMA(即肌成纤维数量)荧光表达情况,PT-PCR和Western blot法检测TGF-β1信号通路情况,来测定紫杉醇对成纤维细胞肌成纤维化的抵抗作用及其最佳抑制浓度和最佳抑制时间。并在以上基础上分析紫杉醇缓释膜的疗效。结果:紫杉醇可通过阻滞TGFβ/Smad信号通路进而抑制胆管瘢痕成纤维细胞向肌成纤维转化,其最佳浓度为0.06 g/m L,最佳时间为48 h;紫杉醇给药时,使用缓释膜释放比直接药物操作更加简便,效果更理想,更能减轻患者的痛苦(P<0.05);紫杉醇-壳寡糖缓释膜抗良性胆管瘢痕纤维化的最佳载药量为9mg。结论:紫杉醇-壳寡糖能够通过抑制TGFβ-Smad信号通路对良性胆管瘢痕纤维化起到抵抗作用,为临床防治良性胆管瘢痕纤维化的研究提供了新的方向,具有较高的临床价值和研究意义。Objective: To investigate the paclitaxel- chitosan sustained-release membrane through the mechanism of inhibition of TGF β/Smad signal the resistance of benign biliary scar fibrosis and its best effect factors. Methods: the extraction of fibroblasts in New Zealand rabbits(FB)were cultured in vitro, respectively adding paclitaxel solution of different concentration, the growth activity of FB cells by the method of MTT, IFCC determination of α-SMA(i.e., muscle fiber number)fluorescence expression, RT-PCR and Western blot detection of TGF-β1 signal pathway, for the determination of paclitaxel on FB muscle resistance fibrosis and best inhibitory concentration and the best inhibition time. The effect and analysis of paclitaxel release film on the basis of the above.Results: paclitaxel can block the TGF β/Smad signaling pathway and inhibition of biliary tract scar fibroblast fibers into muscle, the optimum concentration was 0.06g/m L, the best time was 48 h, paclitaxel administration, using release film release than direct drug operation more convenient, the effect was more ideal, more could alleviate the pain of patients. Conclusion: paclitaxel-chitosan oligosaccharide can through inhibition of TGF β-Smad signaling pathway to resist the role of benign biliary scar fibrosis, provide a new direction for the clinical treatment of benign biliary scar fibrosis,clinical value and significance of the study with hig
