摘 要:目的:探讨山药多糖联合维生素C对镉性小鼠肝损伤的预防性保护作用。方法:将50只昆明种小鼠随机分为5组,即对照组、模型组(常规饲料饲养)、山药多糖组(山药多糖5.0 g/kg饲料)、维生素C组(维生素C 1.5g/kg饲料)、联合组(山药多糖5.0 g/kg+维生素C 1.5 g/kg饲料)。饲养7 d后,除对照组腹腔注射等体积的0.9%氯化钠溶液外,其余各组腹腔注射Cd Cl2(3.0 mg/kg)溶液。第8天取血测ALT和AST。计算小鼠肝体指数(肝脏质量/体质量),用肝匀浆对SOD、GSH-Px、MDA、NO定性定量。结果:与模型组相比,山药多糖、维生素C组肝体指数、血清ALT和AST活性、MDA和NO含量均下降,SOD、GSH-Px活性和GSH含量均升高(P<0.05,P<0.01);山药多糖和维生素C联合应用保护作用更明显(P<0.05)。结论:山药多糖、维生素C可通过拮抗镉所致的毒性和氧化应激,预防性保护镉性小鼠肝损伤,二者联合使用具有协同效应。Objective To investigate the protective effects of yam polysaccharide and vitamin C on cadmium-induced liver injury in mice. Methods A total of 50 Kunming mice were randomly divided into control group,model group( fed with normal diet),yam polysaccharide group( yam polysaccharide 5. 0 g / kg diet),vitamin C group( vitamin C 1. 5 g / kg diet),combination group( yam polysaccharide 5. 0 g / kg and vitamin C 1. 5 g / kg diet). After feding for 7 days,the control group were intraperitoneally injected with 0. 9% sodium chloride saline and the other groups were intraperitoneally injected with Cd Cl2 solution with a dose of 3. 0 mg / kg. Then the mice were killed 24 h after treatment,the ALT and AST levels were detected in the serum of mice,and liver-body mass index were analyzed. Liver homogenate were prepared and the concentrations of MDA,GSH,NO and the activity of SOD,GSH-Px were tested. Results Compared with model group,the level of liver body mass index,the activity of serum ALT,AST,and the concentrations of MDA and NO in liver were all significantly decreased,the content of GSH and the activity of SOD and GSH-Px were all significantly increased in both yam polysaccharide group and vitamin C groups( P〈 0. 05,P〈 0. 01). The most significant changes in above indexes were found in combination group( P〈 0. 05). Conclusion Both yam polysaccharide and vitamin C could prevent liver injury from cadmium-induced toxicity and oxidative stress,the combination of yam polysaccharide and vitamin C showed a synergistic effect.
