健脾补肾方对BALB/c鼠大肠癌肝转移模型中VEGF表达的影响

摘　　要：目的:观察健脾补肾方是否对BALB/c鼠结直肠癌肝转移模型VEGF表达产生影响。方法:将40只大肠癌肝转移的BALB/c鼠随机区组分为生理盐水组、健脾补肾方高剂量组、健脾补肾方中剂量组、健脾补肾方低剂量组4组。健脾补肾方高剂量组43.7 g/(kg·d),健脾补肾方中剂量组21.85 g/(kg·d),健脾补肾方低剂量组10.925 g/(kg·d),对照组则予以生理盐水,4组均为0.2 m L/只,每天灌胃1次,共2周。根据各组HE染色观察肝脏的转移情况、免疫组化和Western blot测量每组的VEGF的表达。结果:HE染色观察:健脾补肾方中、高剂组肿瘤肝转移面积明显小于对照组,免疫组化及Western blot结果均表明健脾补肾方可以下调VEGF蛋白的表达水平(P<0.05)。结论:健脾补肾方可改善BALB/c鼠大肠癌肝转移的情况,可能通过降低VEGF的表达。Objective：To study the effect of Jian Pi Bu Shen Fang（JPBSF）on the expression of VEGF in the BALB/c mouse with the model of CRC liver metastases. Methods：40 BALB/c mice cecal carcinoma in modeling,after modeling they were randomly Randomized blocked into the control group（0.9% normal saline）,high dose group of JPBSF（43.7 g/kg）,medium dose group（21.85 g/kg）and low dosage groups（10.925 g/kg）.Every group has 10 mice. They were intragastric feed once every day,continuous administration of two weeks.Describe the liver metastases by HE staining and detect the expression of VEGF through immunohistochemistry and Western Blot. Results：The JPBSF groups obviously reduced the degree of liver metastases in high dose group and medium dose of JPBSF group compared with the control group by HE staining. Immunohistochemistry and Western blot results revealed that all groups of TCM reduce the expression of VEGF protein by JPBSF. Conclusion：JPBSF can improve the situation of liver metastases with of colorectal cancer in BALB/c mice,perhaps by the way to download the expression of VEGF.
