摘 要:目的:采用星点设计—效应面法优化黄芩中黄酮苷类成分的提取工艺。方法:采用UPLC法测定黄芩苷及汉黄芩苷含量,以二者的总评"归一值(OD)"为因变量,乙醇浓度、提取时间、料液比为自变量,对结果进行多元线性回归和二项式方程拟合,采用效应面法优化提取工艺,并进行预测分析。结果:最佳提取工艺为58%乙醇,超声提取45min,料液比为1∶40,OD预测值(0.997)与验证值(0.991)的偏差小于1%。结论:星点设计—效应面法优选的黄芩中黄酮苷类成分的超声提取工艺简便,模型预测性良好,可为探讨黄芩中苷类成分提取工艺的研究提供参考。Objective:To optimize ultrasonic extraction Technology of baicalin and wogonoside in Scutellaria baicalensis by central composite design-reponse surface method(CCD-RSM).Methods:The contents of baicalin and wogonoside were determined by Ultra performance liquid chromatography(UPLC).Independent variables were ethanol concentration,liquid-solid ratio and extraction time,the dependent variable was the overall desirability of extraction rate of baicalin and wogonoside.Linear or no-linear mathematic models were established to estimate the relationship between independent and dependent variables.Response surface methodology was used to optimize the process of extraction.Prediction was carried out through comparing the observed and predicted values.Results:The optimum extraction technology was as following:The extraction time was 45 min with solvent to solid ratios(v/w)of 40:1,and the extraction solvent was 58% ethanol at the extraction temperature of 40℃.The deviation between the observed and predicted values was small(P〈0.01).Conclusion:This optimized extraction technology was stable and feasible with high predictability.This method can be provided reference for exploring the flavonoid glycosides in Scutellaria baicalensis.
