摘  要:目的:观察补肾化痰益智法对APP/PS1双转基因小鼠学习记忆及抗氧化能力的影响并探讨其可能机制。方法:随机将APP/PS1双转基因小鼠分为模型组、西药多奈哌齐组(6.5×10-4g·kg-1·d-1)、补肾化痰益智方高、低剂量组(15.6,7.8 g·kg-1·d-1),每组10只。空白组为10只同月龄相同遗传背景C57BL/6J小鼠。从3月龄开始对小鼠分别实施灌胃治疗,每日1次。空白组、模型组均给予等容积的生理盐水灌胃。连续灌胃3个月后,进行跳台实验及穿梭箱实验评定各组小鼠的学习记忆能力;采用比色法检测小鼠脑组织超氧化物歧化酶(SOD)和丙二醛(MDA)的含量。结果:跳台实验结果显示,与模型组比较,各组小鼠的潜伏期明显延长和出错次数明显减少(P<0.01);穿梭箱实验结果显示,与模型组比较,各组小鼠的主动回避次数明显增加(P<0.01,P<0.05),主动回避潜伏期、被动回避潜伏期明显缩短(P<0.01);西药组和补肾化痰益智方高、低剂量组小鼠脑组织中SOD活性较模型组显著升高(P<0.01),MDA含量明显降低(P<0.01)。结论:补肾化痰益智方对APP/PS1双转基因小鼠的学习记忆具有一定的改善作用,且可能与其抗氧化作用有关。Objective: To investigate the effect of replenishing kidney-essence and removing phlegm on the ability of learning and memory and antioxidant in APP/PS1 double transgenic mice. Method: APP/PS1 double mice were randomly divided into model group, western medicine group (6.5×10^-4g·kg^-1·d^-1), Bushen Huatan Yizhi recipe (BSHTYZR) high dose group (15.6,7.8 g·kg^-1·d^-1 ), low dose group (7.8 g·kg^-1·d^-1) , 10 in each groups. Blank group mice (C57BL/6J) were 10 months of age with the same genetic background mice. After intragastrical administration for 3 months, the ability of learning and memory was assessed by the step down avoidance test and shuttle box test; colorimetric assay was used to measure mouse brain tissue superoxide dismutase (SOD) and malondialdehyde (MDA) content. Result: Step down avoidance test results showed that compared with model group, each group of mice significantly prolonged the incubation period and the number of errors decreased significantly (P 〈 0.01 ) ; shuttle box test results showed that compared with model group, the number of active avoidance in treatment groups increased significantly ( P 〈 0.01, P 〈 0.05 ) , and active avoidance incubation period, the passive avoidance period significantly shorten (P 〈 0.01 ) ; shuttle box test results showed that compared with the model group, in the rest of the administration group the small active avoidance significantly increased the number of rats ( P 〈 0.01, P 〈 0.05 ) , while the active avoidance latency and passive avoidance latency was shorter (P 〈 0.01 ) ; in addition, in BSHTYZR high dose group and low dose group SOD activity of brain tissue increased significantly compared with the model group (P 〈 0.01 ), MDA was significantly lower than the model group (P 〈0.01 ). Conclusion: BSHTYZR has a certain improvement on APP/ PS1 double transgenic mice learning and memory, and may be related to the antioxidant effects.
