摘 要:目的:观察涤痰汤对老年轻度认知功能障碍(MCI)大鼠海马β淀粉样蛋白(Aβ1-42)含量及早老素1(PS-1)、早老素2(PS-2)表达的影响。方法:将60只13月龄SD大鼠随机分为正常组、模型组、西药组、涤痰汤高剂量组、涤痰汤低剂量组,另设12只3月龄青年大鼠作为青年对照组。模型组、西药组、涤痰汤高、低剂量组采用中年大鼠颈部皮下注射D-半乳糖8周结合半高脂饮食4周的方法建立老年轻度认知功能障碍大鼠模型。第5-8周,涤痰汤高、低剂量组分别ig给予涤痰汤8.55,4.275 g·kg-1,西药组ig给予盐酸多奈哌齐0.9 mg·kg-1,空白组、模型组、青年组ig给予等容积的蒸馏水,每日1次。采用Morris水迷宫检测大鼠学习记忆能力,酶联免疫测定法(ELISA)检测大鼠海马组织Aβ1-42含量,免疫组化法检测海马PS-1,PS-2阳性表达。结果:与空白组比较,模型组大鼠平台逃避潜伏期延长(P〈0.01),海马CA1区PS-1,CA3区PS-2阳性表达增多(P〈0.01),海马Aβ1-42含量上升(P〈0.05)。与模型组相比,涤痰汤低、高剂量组大鼠平台逃避潜伏期缩短(P〈0.05,P〈0.01),海马Aβ1-42含量下降(P〈0.05,P〈0.01),海马PS-1,PS-2表达均下降(P〈0.01)。结论:涤痰汤可能通过下调模型大鼠海马PS-1,PS-2水平,降低海马Aβ1-42含量,从而改善老年认知功能障碍大鼠的学习记忆能力。Objective: To observe the influence of Ditan Tang on the content of/3-amyloid (Aβ1-42) and expressions of presinilin (PS) -1, PS-2 in hippocampus of senile mild cognitive impairment (MCI) rats. Method: Sixty 13 months old rats were randomly divided into normal group, model group, Donepezil 0. 9 mg · kg^-1 group, the high dose and low dose groups (8.55, 4.28 g · kg^-1) of Ditan Tang, 12 three months old rats were as young normal group. The senile MCI rats model was established by subcutaneous injection of D-galactose for 8 weeks and fed with semi-high-fat diet for 4 weeks in model group, Donepezil group, the high dose Ditan Tang group, and low dose Ditan Tang group. The learning and memory ability was tested by Morris water maze test in rats. Enzyme linked immunosorbant assay (ELISA) method was used to detect the levels of Aβ1-42in hippocampus. Levels of PS-1 and PS-2 in hippocampus were detected by immunohistochemistry. Result: Compared with normal group, the escape latency in model group rats was significantly prolonged (P 〈 0.01) , the content of Aβ1-42 in hippocampus increased significantly (P 〈 0.01 ) , and the expressions of PS-1 and PS-2 in hippocampus also increased (P 〈 0.05). Compared with model group, the escape latency of Ditan Tang group (high dose and low dose) were significantly shorten (P 〈 0.01, P 〈 0.05). The content of Aβ1-2 in hippocampus on both high and low dose group of Ditan Tang decreased (P 〈 0.01, P 〈 0.05). The expressions of PS-1 and PS-2 in hippocampus on both high and low dose group decreased significantly (P 〈 0.01 ). Conclusion: The Ditan Tang may decrease the contents of Aβ1-42 and the expressions of PS-1 and PS-2 in hippocampus. It might be reason of improving cognitive dysfunction in rats.
