摘 要:目的观察归芷软胶囊(白芷、当归、珍珠粉、银杏叶等)对黄褐斑模型小鼠肝脏和皮肤中酪氨酸酶水平、超氧化物歧化酶(SOD)活力、丙二醛水平、皮肤中黑色素细胞免疫组化的影响。方法 60只SPF级雌性ICR小鼠随机均分成空白组,模型组,VC片组,归芷软胶囊低、中、高剂量,除空白组外各组接受中波紫外线照射加肌肉注射黄体酮诱发黄褐斑。测定各组小鼠肝脏和皮肤中的酪氨酸酶和丙二醛的水平,SOD活力,同时采用免疫组化法观察小鼠皮肤中Melan A的表达。结果与模型组相比,归芷软胶囊低、中、高剂量组肝脏和皮肤中酪氨酸酶和丙二醛水平,皮肤中黑色素细胞的阳性目标面积、平均直径、积分光密度明显降低,有显著性差异(P<0.01或P<0.05);阳性对照组维生素C片组小鼠皮肤中SOD活力明显升高,有显著差异(P<0.05),其它组没有显著性差异。结论归芷软胶囊对黄褐斑模型小鼠有一定的防治作用。其机制可能与抑制酪氨酸酶活性和黑色素细胞的增殖,提高机体抗氧化能力有关。AIM To observe the impacts of Guizhi Soft Capsules( Angelicae dahuricae Radix,Angelicae sinensis Radix,Pulvis Pernnlae,Ginko Folium,etc) on tyrosinase level,superoxide dismutase( SOD) activity,malondialdehyde( MDA) level in the liver and skin of mouse model for chloasma. METHODS Sixty ICR female mice of SPF grade were equally assigned into blank,model,Vitamin C,Guizhi Soft Capsules low-,middle-,and high-dose groups. Except the blank group,all of the rest animals received ultraviolet irradiation and intramuscular injection of progesterone,which induced chloasma. After the treatment,the liver and skin were collected,their tyrosinase level,SOD activity,malondialdehyde level were detected and Melan A on skin was observed by immunohistochemistry. RESULTS Compared with the model group,the tyrosinase and malondialdehyde levels in mouse skin and liver tissue,and the target skin area,the diameter( mean),the integrated optical density in the skin tissue of Guizhi Soft Capsules low-,middle-,and high-dose groups decreased significantly( P〈 0. 01 or P〈 0. 05). The SOD activity in mouse skin tissue of the Vc group increased significantly( P〈 0. 05). No significant differences were observed in other groups. CONCLUSION Guizhi Soft Capsules plays a significant role in the protective and treatment of the mouse model for chloasma. The mechanism of the action may be related to inhibiting the activity of tyrosinase and the generation of skin melanoma cell,and thus improves the oxidation resistance of tissues.
