摘 要:【目的】探讨烧伤后小鼠血清皮质醇及肝组织皮质醇结合球蛋白(corticosterone binding globulin,CBG)的时相变化特点。【方法】将40只C57BL/6小鼠随机分为正常对照组和实验组,实验组造成Ⅲ度烧伤,烧伤面积15%~20%。分别于伤后12、24、48 h测定血清总皮质醇、游离皮质醇浓度,采用免疫印迹、逆转录PCR在蛋白质和m RNA水平测定肝组织CBG水平。【结果】烧伤后小鼠血清总皮质醇浓度升高,12 h升至顶峰,为(95.52±6.73)mmol/L,48 h恢复正常,为(50.99±9.52)mmol/L;同时肝组织CBG m RNA和CBG水平下降,使血清游离皮质醇浓度升高,烧伤后24 h升至顶峰,为(25.6±5.01)mmol/L,48 h时仍未恢复正常,为(16.06±6.56)mmol/L。【结论】烧伤应激引起小鼠血清皮质醇水平明显升高,CBG水平下降对血清皮质醇起正相调节作用。[ Objective]To observe characteristics of the phase changes of the mice serum cortisol and liver tissue Corticosterone Bind- ing Globulin(CBG) levels after burn. [ Method ] A total of 40 C57BL/6 mice were dividedrandomly into control groups(n = 10) and burn groups (n = no, respectively). After shaving, the mice in burn groups were inflicted with 15%--20%, TBSA Ⅲ degree. Serum total eorti- sol and free cortisol concentrations were measured while the CBG protein levels in mice liver tissue were measured by Western blotting and the CBG mRNA levels were measured by RT-PCR in 12, 24 and 48 h after burn respectively. [ Result] Serum total cortisol concentrations of burn groups were elevated to peak in 12 h[(95.52 ± 6.73) mmol/L] and returned to normal in 48 h [(50.99 ± 9.52) mmol/L], while free cortisol was continuously elevated to peak in 24 h[(25.6 ± 5.01) mmol/L] and not returned to normal until 48 h[(16.06 ± 6.56) mmol/L] with the low expression of CBG and CBG mRNA in liver tissue. [Conclusion] Burn stress caused serum cortisol levels to increase significantly. The decline of CBG and CBG mRNA levels in liver played a positive phase regulatory role on serum cortisol after burn.
