摘  要:目的:探讨大肠杆菌疫苗对发育期胎兔肺表面活性蛋白(SPs)基因表达的影响。方法:将孕兔30只随机分为大肠杆菌疫苗组、地塞米松组和生理盐水组各10只。大肠杆菌疫苗组于妊娠第10、17、24天宫颈肌肉内注射大肠杆菌疫苗0.1 ml/kg,地塞米松组和生理盐水组妊娠第22、23、24天分别肌肉注射地塞米松2.0 mg/kg、生理盐水2.0 mg/kg。于妊振第27天剖宫产取出胎兔肺,逆转录PCR(RT-PCR)观察分析各组胎兔肺表面活性蛋白SP-A、SP-B和SP-C mRNA表达变化,以B-actin mRNA扩增产物为内参照行半定量分析。结果:在第27天时,生理盐水组胎兔肺SP-A、SP-B和SP-C mRNA的相对值分别为0.83±0.26、0.97±0.18、0.88±0.11,大肠杆菌疫苗组胎兔肺SP-A、SP-B mRNA的相对值分别是0.98±0.17、1.12±0.23,两组比较差异有统计学意义(P<0.05),SP-C mRNA的相对值是0.90±0.12,两组比较差异无统计学意义(P>0.05);地塞米松组胎兔肺SP-A、SP-B和SP-C mRNA的相对值分别为1.08±0.21、1.14±0.27、1.02±0.25,与生理盐水组比较差异有统计学意义(P<0.05)。结论:地塞米松对胎兔肺SPs基因表达有显著的促进作用,大肠杆菌疫苗对胎兔肺SPs基因表达也有一定促进作用。Objective: To explore the effect of Escherichia Coli vaccine on gene expression of pulmonary surfactant proteins( SPs)in fetal rabbit lungs. Methods: Thirty pregnant rabbits were randomly divided into Escherichia Coli vaccine group,dexamethasone group,and normal saline group,ten rabbits in each group. The rabbits in Escherichia Coli vaccine group were treated with intramuscular injection of Escherichia Coli vaccine( 0. 1 ml / kg) on the 10 th,17th,and 24 th days during pregnancy; the rabbits in dexamethasone group were treated with intramuscular injection of dexamethasone( 2. 0 mg / kg) on the 22 th,23th,and 24 th days during pregnancy; the rabbits in normal saline group were treated with intramuscular injection of normal saline( 2. 0 mg / kg) on the 22 th,23th,and 24 th days during pregnancy. RT-PCR was used to observe the changes of SP- A,SP- B,and SP- C mRNA expressions in the three groups; semiquantitative analysis was conducted taking B- actin mRNA amplification product as internal reference. Results: On the 27 th day during pregnancy,the relative values of SP- A,SP- B,and SP- C mRNA in normal saline group were 0. 83 ± 0. 26,0. 97 ± 0. 18,and 0. 88 ± 0. 11,respectively; the relative values of SP- A and SP- B mRNA in Escherichia Coli vaccine group were 0. 98 ± 0. 17 and 1. 12 ± 0. 23,respectively,there were statistically significant differences in the relative values of SP- A and SP- B mRNA between the two groups( P 〈0. 05),the relative value of SP- C mRNA in Escherichia Coli vaccine group was 0. 90 ± 0. 12,compared with normal saline group,there was no statistically significant difference( P 〉0. 05); the relative values of SP- A,SP- B,and SP- C mRNA in dexamethasone group were 1. 08 ± 0. 21,1. 14 ± 0. 27,and 1. 02 ± 0. 25,respectively,compared with normal saline group,there were statistically significant differences( P 〈0. 05). Conclusion: Dexamethasone can significantly promote SPs gene expressions in fetal rabbit lungs,Escherichia Coli vaccine can promote
