摘  要:为了探究移植脐带间充质干细胞(UC-MSCs)对荷斯坦公犊牛日增重和血清中激素水平的影响,试验选取1月龄体重相近的荷斯坦公犊牛28头,随机分为对照组和试验组;采集健康的荷斯坦新生胎牛的脐带组织,通过胰酶消化筛选法体外分离、纯化及培养牛脐带间充质干细胞(UCMSCs),试验组公犊牛注射干细胞3×105个/kg(稀释至8 m L颈静脉注射),对照组注射等量生理盐水,试验期为155 d。结果表明:注射UC-MSCs后,4,5,6月龄时,试验组公犊牛平均体重显著高于对照组(P<0.05)。4月龄时,试验组公犊牛平均日增重显著高于对照组(P<0.05);6月龄时,试验组公犊牛平均日增重极显著高于对照组(P<0.01)。4月龄时,试验组公犊牛血清中生长抑素(SS)水平显著低于对照组(P<0.05);6月龄时,试验组公犊牛血清中甲状腺素(T4)、三碘甲腺原氨酸(T3)水平差异极显著(P<0.01)。说明移植UC-MSCs能显著增加荷斯坦公犊牛的体增重,降低血清中生长激素(GH)、SS、瘦素(LEP)的水平,提高血清中T4、T3水平,而血清中这些激素水平与公犊牛的生长发育密切相关,进而影响其生长性能。To explore the effect of the transplantation of umbilical cord mesenchymal stem cells( UC- MSCs) on the daily gain and serum hormone levels in Holstein bull calves,28 Holstein calves with similar body weight at one month of age were selected,and then were randomly divided into the control and experimental groups. The umbilical cord tissues from healthy Holstein fetal calves were collected and used to purify and cultivate the bovine UC- MSCs in vitro using the trypsin digestion method. The bull calves from the experimental group were injected in jugular vein with the UC- MSCs of 3 × 10^5 cells / per kg which were diluted to 8m L,and the control group was injected with saline. The test lasted for 155 d. The results showed that after the injection of UC- MSCs,the average body weight in the experimental group was significantly higher( P〈0. 05) than that in the control group at four,five and six months of age; the average daily gain in the experimental group was significantly higher( P〈0. 05) than that in the control group at four months of age; the average daily gain in the experimental group was extremely significantly higher( P〈0. 01) than that in the control group at six months of age. The serum somatostatin( SS) levels in the experimental group were significantly lower( P〈0. 05) than those in the control group at four months of age; the serum thyroxine( T4) and triiodothyronine( T3) levels showed extremely significant difference( P〈0. 01) in the experimental group at six months of age. The results indicate that the transplantation of the UC- MSCs can significantly increase the body weight gain of Holstein bull calves and decrease the serum growth hormone( GH),SS,leptin( LEP),and hormone levels,and improve the serum T4 and T3 levels. These serum hormone levels are closely related to the growth and development of Holstein bull calves,thereby affecting their growth performance.
