摘  要:目的研究αB-晶状体蛋白对钾离子通道kv1.1、kv1.3及凋亡抑制因子Bcl-xl和凋亡因子Caspase-3的影响,探讨αB-晶状体蛋白对急性高眼压大鼠视网膜神经节细胞(retinal ganglion cells,RGC)保护作用的机制。方法 60只SD大鼠随机分为A组(急性高眼压组)、B组(急性高眼压+玻璃体内注射生理盐水5μL组)、C组(急性高眼压+玻璃体内注射10×10-6 g·L-1αB-晶状体蛋白5μL组),每组20只,右眼为实验眼。注射后7 d和14 d时,应用Western blot和实时定量逆转录聚合酶链反应(QRT-PCR)法检测kv1.1、kv1.3、Bcl-xl和Caspase-3在视网膜上的表达水平。结果注射后7 d和14 d时,C组kv1.1、kv1.3通道蛋白和凋亡因子Caspase-3蛋白表达水平和mRNA相对表达量均较A组、B组低,差异均有统计学意义(均为P<0.05);C组Bcl-xl蛋白表达水平和mRNA相对表达量较A组、B组高,差异均有统计学意义(均为P<0.05);相同时间点不同因子A组、B组间两两比较,差异均无统计学意义(均为P>0.05)。结论αB-晶状体蛋白通过抑制急性高眼压后RGC上钾离子通道kv1.1、kv1.3的表达,从而提高凋亡抑制因子Bcl-xl的表达、降低凋亡因子Caspase-3的表达,减少细胞凋亡,保护RGC。Objective To investigate the mechanism of αB-crystallin in protecting retinal ganglion cells of rats with acute ocular hypertension through observing the effects of αB-crystallin on the expression of potassium ion channel kv1. 1 and kv1. 3,anti-apoptosis factors Bcl-xl and apoptotic factors Caspase-3. Methods Sixty SD rats were randomized into group A( acute high intraocular pressure group),group B( acute high intraocular pressure following intravitreal injection of normal saline group),group C( acute high intraocular pressure following intravitreal injection of 10 × 10- 6 g·L- 1 αB crystallin group),20 cases in each group,the right eye was the experimental eye. At 7 days and 14 days after injection,the expression of kv1. 1,kv1. 3,Bcl-xl and Caspase-3 in retina were observed by Western blot and QRT-PCR. Results At 7 days and14 days after injection,the relative protein and mRNA expression levels of kv1. 1,kv1. 3 and Caspase-3 in group C were lower than those in group A and B,the difference was significant( all P〈0. 05); The relative protein and mRNA expression levels of Bcl-xl in group C were higher than those in group A and B,the difference was significant( all P〈0. 05); The relative protein and mRNA expression levels of BCL-XL in group C were higher than those in group A and B,the difference was significant( all P〈0. 05). There was no statistical difference in each index at each time point between group A and group B( P〉0. 05). Conclusion αB-crystallin can inhibit the expression of potassium ion channels kv1. 1,kv1. 3 in the retinal ganglion cells after acute high intraocular pressure,so it can increase the expression of anti-apoptosis factors Bcl-xl anddecrease the expression of apoptotic factors Caspase-3,then reduce apoptosis and protect RGC.
