Wortmannin对糖尿病肾病大鼠肾组织IL-1与IL-6表达的影响

摘　　要：目的研究糖尿病肾病(DN)大鼠模型经静脉注射PI3K抑制剂Wortmannin后肾组织白介素-1(IL-1)与白介素-6(IL-6)表达的变化。方法成年健康清洁级SD雄性大鼠30只,随机分为假手术组(10)、模型组组(10)和Wortmannin处理组(10)。参照Anderson's方法制作糖尿病肾病大鼠模型。采用Western blot方法检测三组大鼠肾组织IL-1与IL-6蛋白的表达;RT-PCR方法检测三组大鼠肾组织IL-1m RNA与IL-6m RNA的表达。结果模型组与Wortmannin处理组肾组织IL-1与IL-6蛋白和m RNA的表达量均显著高于假手术组(<0.05),而与模型组相比,Wortmannin处理组肾组织IL-1与IL-6蛋白和m RNA的表达量明显降低(<0.05)。结论 PI3K抑制剂Wortmannin能够下调DN大鼠肾组织IL-1与IL-6的表达。Objective To study the effect of Wortmanninon on expressions of interleukin-1（IL-1） and Interleukin-6（IL-6） in kidney of the diabetic nephopathy rat. Methods 30 healthy male rats were selected and randomly divided into the sham group（10）, the model group（10）and Wortmannin treatment group. The diabetic nephropathy model was induced with Anderson＇ s way. The expressions of IL-1 and IL-6 protein and m RNA were observed by Western blot and RT-PCR respectively. Results The expression levels of IL-1 and IL-6 protein and m RNA were increased significantly in model group and Wortmannin treatment group than in sham group（ P〈0.05）, but decreased in Wortmannin treatment group than in model group（P〈 0.05）. Conclusion Wortmannin could down-regulate the expressions of IL-1 and IL-6 in diabetic nephropathy rat kidney.
