摘 要:目的建立环磷酰胺致免疫低下模型,评价不同剂量环磷酰胺对正常小鼠免疫抑制的作用效果,探究环磷酰胺免疫抑制小鼠的适宜剂量。方法采取不同时间给予正常小鼠腹腔注射不同剂量(40,80,100mg·kg^(-1)·d^(-1))的环磷酰胺,分别建立免疫低下模型,通过免疫脏器指数、小鼠腹腔巨噬细胞吞噬鸡红细胞、CCK-8法检测淋巴细胞增殖、ELISA法检测血清中IgM的表达水平探讨环磷酰胺对小鼠免疫功能的影响。结果腹腔注射环磷酰胺3个剂量组(40,80,100mg·kg^(-1)·d^(-1))的正常小鼠均能不同程度表现免疫脏器指数下降、单核巨噬细胞吞噬能力降低、淋巴细胞增殖能力及小鼠血清溶血素表达水平低下现象。结论正常小鼠腹腔注射不同浓度的环磷酰胺(40,80,100mg·kg^(-1)·d^(-1)),可建立免疫抑制模型,并且连续3d腹腔注射剂量为80mg·kg^(-1)·d^(-1)的环磷酰胺可以显著降低小鼠免疫功能,其效果最明显,适用于建立免疫低下模型。[ ABSTRACT] Objective To establish cyclophosphamide-induced immunocompromised model ,evaluate inhibited effects of different doses of cyclophosphamide on mice immunity and investigate the right dose .Methods Different doses of cyclophosphamide were injected at different times respectively to establish immunosuppressive models ,immune organ index,mononuclear macrophage ,lymphocyte proliferation tested by CCK-8,serum hemolysin formation examined by ELISA were used to explore the effects of cyclophosphamide on mouse immune function .Results Three different doses of cyclophosphamide all lead to the decrease of immune organ index ,mononuclear macrophage phago-cytosis,lymphocyte proliferation and serum hemolysin expression .Conclusion Different doses of cyclophosphamide can be injected to esta-bilish immunosuppression modes and continuous intraperitoneal injection of 80mg/kg/day of cyclophosphamide for 3 days can significantly re-duce the immune function in mice .Because of its significant effect ,it is suitable for the establishment of long-term immunosuppression mod-els.
