摘  要:目的研究AG490对大鼠创伤性脑损伤后脑组织CD11b表达及血脑屏障(BBB)通透性的影响。方法取健康成年雄性SD大鼠72只,随机分为空白组、创伤组、AG490干预组,每组24只,每组又分为4个亚组,分别对应伤后4h,1d,3d,7d时间点,每亚组6只,应用液压冲击法制作大鼠脑创伤模型,用伊文思蓝渗透法检测BBB通透性改变情况;应用免疫组化法检测脑组织中CD11b表达水平。结果脑损伤后,BBB通透性增加,与空白组比较,创伤组伤区脑组织伊文思蓝渗透量均增加(P<0.01),伤后1d升高,3d维持高值状态,然后逐渐下降。AG490干预组各时间段均比创伤组伊文思蓝渗透量低(P<0.01);免疫组化显示CD11b表达阳性细胞镜下呈棕黄色颗粒状,脑创伤后CD11b在创伤组各时间段表达量均明显高于对照组(P<0.01),AG490干预组各时间段表达量低于创伤组(P<0.01)。结论脑损伤后急性期应用AG490可降低创伤性脑损伤后小胶质细胞活化程度,减轻BBB的破坏,有助于减轻继发性脑损伤。Objective To investigate the effect of AG490 on the brain barrier permeability and the expression of CD 11b after traumatic brain injury in rats .Methods A total of 72 healthy male SD rats were randomly divided into a sham group ,a TBI group and an AG490 intervention group .The rats in each group were redivided into 4h,1d,3d,7d subgroups according to the time of brain injury .A brain trauma model was induced by using a hydraulic shock method;The rats in each group were redivided into 4h,1d,3d,7d subgroups according to the time of brain injury and the brain injury models were made by hydraulic impact .The BBB permeability was determined by Evan's blue (EB) extravasation method;Immunohistochemistry was used to determine the expression of CD 11b in injured brain tissue.Results The per-meability of BBB increased after traumatic brain injury ,the Evans blue permeation of all time points in the traumatic brain injury group were higher than those in the sham group ,it began increase at 1d,it maintain a high value status at 3d,and then gradually downward .The Evans blue permeation in the AG490 intervention group were lower than those in the traumatic brain injury group (P〈0.01);Immunohistochemistry showed CD11b positive cells were showed brown granular under microscope ,the expression levels of CD11b in the traumatic brain injury group was higher than that in the sham group at each time point (P〈0.01).Compared with the traumatic brain injury group ,the expression levels of CD11b in the AG490 intervention group were lower (P〈0.01).Conclusion Applying of AG490 after the acute phase of traumatic brain injury can reduce the activation of microglial cells ,reduce the blood-brain barrier damage and help to reduce secondary brain injury .
