摘 要:目的探讨金黄色葡萄球菌肠毒素B(SEB)对于皮炎局部糖皮质激素受体β(GRβ)的影响。方法 1%DNCB重复刺激Balb/c小鼠背部皮肤构建特应性皮炎动物模型,以未经处理的模型小鼠为对照组,并分别用SEB、SEB+地塞米松、地塞米松处理该动物模型。通过H&E染色观察组织病理变化,计数真皮内炎性细胞数量来反映不同处理组皮炎的变化情况,采用q RT-PCR、Western blot和免疫组织化学方法检测糖皮质激素受体β(GRβ)的m RNA和蛋白表达。结果在利用DNCB构建的Balb/c小鼠特应性皮炎动物模型中,真皮内炎性细胞数量明显增加;与未经处理的皮炎组相比,SEB处理后炎性细胞显著增加,地塞米松处理后炎性细胞显著减少,但SEB可部分拮抗地塞米松的抗炎作用;此外,在SEB处理的皮炎组以及SEB+地塞米松处理的皮炎组,GRβm RNA和蛋白的表达均显著上调,但SEB单独处理的皮炎组GRβm RNA和蛋白的表达上调情况更为显著。结论细菌超抗原(SEB)在皮炎局部诱导了GRβ的表达升高,在外用糖皮质激素快速减敏过程中起到了重要的作用。The aim of this study is to investigate the possible involvement of staphylococcal enterotoxin B(SEB) in glucocorticoid tachyphylaxis and the molecular mechanisms. Atopic dermatitis animal model was established by repetitive stimulation of 1% DNCB on the Balb/c mouse dorsal skin, taking the untreated mouse model as control group, and then respectively administrate SEB, SEB+dexamethasone, dexamethasone on the model.The histopathology changes were observed by HE staining; the number of the inflammatory cells in dermal was counted for evaluated the dermatitis change of different treatment groups; the m RNA and protein expression of glucocorticoid receptors β(GRβ) were detected by adopting the methods of q RT-PCR, Western blot and immunohistochemistry. Data showed that the number of the inflammatory cells in dermal was significantly increased in the atopic dermatitis model. Compared with the untreated atopic dermatitis group, SEB significantly increased the number of inflammatory cells, while dexamethasone significantly decreased the number of inflammatory cells. And SEB has partial antagonism on anti-inflammatory effect of dexamethasone. Additionally, as to the dermatitis group treated with SEB and the dermatitis group treated both with SEB and dexamethasone, the expression of GRβ m RNA and protein were both significantly up-regulated, while the elevation of GRβ m RNA and protein expression was much higher in the dermatitis group only treated with SEB. Finally draw the conclusion that the rising expression of GRβ in dermatitis induced by bacterial superantigen(SEB) plays an important role in the process of topical glucocorticoid tachyphylaxis.
