摘 要:目的:确定人参皂苷Rg1对UVB损伤成纤维细胞的保护作用。方法:体外培养人皮肤成纤维细胞,分为正常对照组、UVB照射组、UVB照射+人参皂苷Rg1组(5,10和20μg/mL),MTT法检测细胞增殖活性,ELISA法检测细胞中SOD活性、MDA和细胞上清液中MMP-1和MMP-3的含量,Western blot法检查细胞中MMP-1和MMP-3的表达。结果:UVB联合不同浓度人参皂苷Rg1组较UVB照射组,细胞SOD活性升高,MDA、MMP-1和-3含量均有不同程度下降,人参皂苷Rg1浓度越高作用越明显。结论:人参皂苷Rg1对UVB造成的皮肤成纤维细胞损伤具有保护作用。Objective:To determine the protective effects of ginsenoside Rg1 against UVB-induced photo-damage on skin fibroblasts. Methods:Human skin fibroblasts cultured in vitro were divided into normal control group, UVB group, UVB combined ginsenoside Rg1 groups (concentration of 5, 10 and 20 μ g/mL). The proliferation activity of cells was detected by MTT assay. SOD activity, MDA and the content of MMP-1 and MMP-3 were detected by ELISA. The expression of MMP-1 and MMP-3 proteins was detected through Western blot. Results:The SOD activity in UVB combined ginsenoside Rg1 groups was higher than that in UVB group. The content of MMP-1 and MMP-3 in UVB combined ginsenoside Rg1 groups was lower than that in UVB group. The effect was dose dependent. Conclusion: Ginsenoside Rg1 can protect the dermal fi-broblasts from UVB damage.
