摘 要:应用SRAP分子标记引物对24份桑树品种资源的基因组DNA进行PCR,检测其遗传多样性,分析品种间的亲缘关系。15对SRAP引物扩增产生126条谱带,平均每对引物产生8.4条谱带,其中多态性条带共99条,多态性比率为78.57%,说明24个桑树品种间具有丰富的遗传多样性。基于品种间遗传相似系数的UPGMA聚类图和基于品种的SRAP标记类型的主成分分析中,蒙桑种(Morus mongolica Schneid.)的品种吉蒙桑、白桑种(Morus alba L.)的品种垂枝桑和山桑种(Morus bombycis Koidz.)的品种昌农山桑单独聚类,其他来自白桑种、广东桑种(Morus atropurpurea Roxb.)、鲁桑种(Morus multicaulis Perr.)的栽培品种分为3组;白桑种种内各品种间的遗传变异较小,而广东桑种、鲁桑种种内各品种间的遗传变异相对较大;广东桑3个品种间的遗传背景差异较大;鲁桑栽培品种农桑12号与其父本桐乡青的遗传相似度高,而与母本北区一号的差异显著,并且鲁桑种内各品种间未能完全聚类,品种间的亲缘关系与地理分布位置的距离相关。筛选的15对SRAP引物能够有效地检测供试桑树品种的遗传多样性,揭示品种间的亲缘关系。By using SRAP molecular markers to amplify the genomic DNAs of 24 mulberry variety resources,we detected genetic diversity and analyzed genetic relationship between these tested mulberry varieties. Amplification of 15 SRAP primer pairs yielded 126 bands( 8. 4 bands per primer pair),among which 99 bands showed polymorphism with a polymorphic percentage of 78. 57%,implying that there was rich genetic diversity among the 24 tested mulberry varieties. UPGMA dendrogram based on genetic similarity coefficient and principal component analysis based on SRAP marker types showed that Jimeng Sang( Morus mongolica Schneid.),Chuizhi Sang( Morus alba L.) and Changnong Shansang( Morus bombycis Koidz.) formed a monophyletic clade,and the other mulberry cultivars from M. alba,Morus atropurpurea and Morus multicaulis Perr. formed 3 separate groups. The genetic variation level was low among varieties of M. alba species,but was relatively high among varieties of M. atropurpurea Roxb. and M. multicaulis species. Three varieties of M. atropurpurea species had distant genetic background. Nongsang No. 12 had high genetic similarity with the male parent of TongxiangQing,but had significant difference with the female parent of Beiqu No. 1. In addition, different varieties of M.multicaulis species were tangled in different clustering groups,and the genetic relationship between different varieties was correlated with distace of their geographical distribution. These results showed that the screened 15 SRAP primer pairs could effectively detect genetic diversi-ty of the tested mulberry varieties so as to reveal the genetic relationship between different varieties.
