摘  要:本试验通过研究精氨酸(Arg)对氧化应激仔猪内分泌及生长相关因子基因表达的影响,探寻Arg抗氧化应激的作用机制。选择24头健康PIC断奶仔公猪,随机分为4组,每组6个重复,每个重复1头猪。各组试验猪分别饲喂基础饲粮+注射生理盐水(对照组)、基础饲粮+注射Diquat(氧化应激组)、基础饲粮+0.8%Arg+注射Diquat(0.8%Arg组)、基础饲粮+1.6%Arg+注射Diquat(1.6%Arg组)。在试验第8天,各试验组仔猪腹腔注射Diquat溶液(10 mg/kg BW)诱导氧化应激,对照组仔猪注射等量的灭菌生理盐水。正式试验期共11 d。结果表明:1)氧化应激显著降低了仔猪应激后24 h的血浆胰岛素和胰岛素样生长因子-Ⅰ(IGF-Ⅰ)的浓度(P<0.05),补充1.6%Arg显著增加了氧化应激仔猪应激后48、96 h的血浆IGF-Ⅰ浓度(P<0.05)。2)氧化应激对仔猪血浆一氧化氮合成酶的活性无显著影响(P>0.05),显著降低了仔猪肝脏一氧化氮合成酶的活性(P<0.05);补充1.6%Arg可显著抑制氧化应激导致的肝脏一氧化氮合成酶活性的降低(P<0.05)。3)氧化应激可显著降低肝脏IGF-Ⅰ、胰岛素样生长因子-Ⅰ受体(IGF-ⅠR)、胰岛素样生长因子结合蛋白3(IGFBP3)mRNA的相对表达量(P<0.05),而对肌肉中这3个基因mRNA的相对表达量无显著影响(P>0.05);补充1.6%Arg可显著增加肝脏IGF-Ⅰ、IGFBP3及肌肉IGF-Ⅰ、IGF-ⅠR和IGFBP3 mRNA的相对表达量(P<0.05)。综上所述,Arg通过促进胰岛素、IGF-Ⅰ的分泌进而上调生长相关因子基因表达是其缓解仔猪氧化应激的途径之一。The present study was conducted to test the effects of arginine ( Arg) supplementation on endocrine and growth-relative factor gene expression of piglets under oxidative stress, in order to explore the mechanism of Arg on anti-oxidative stress. A total of 24 PIC weaned female piglets were randomly allocated into 4 groups with 6 replicates per group, and each replicate had 1 piglet. Piglets in those groups were fed basal diet+injected sterile saline ( control group ) , basal diet+injected Diquat ( oxidative stress group ) , basal diet supplemented with 0.8% Arg+injected Diquat ( 0. 8% Arg group) , and basal diet supplemented with 1. 6% Arg+injected Diquat (1.6% Arg group), respectively. On day 8 of experiment, piglets were injected intraperitoneally either with diquat ( experiment groups) or sterile saline ( control group) at 10 mg/kg BW to induce oxidative stress. The experiment lasted for 11 d. The results showed as follows:oxidative stress significantly decreased the con-centrations of insulin and insulin-like growth factor-Ⅰ ( IGF-Ⅰ) in plasma at 24 h post-stress of piglets ( P〈0.05) . Supplementation of 1.6% Arg significantly increased the concentration of IGF-Ⅰ in plasma at 48 and 96 h post-stress of piglets under oxidative stress (P〈0.05). 2) Oxidative stress didnot affect the activities of nitric oxide synthases (NOS) in plasma of piglets (P〉0.05). However, Oxidative stress significantly de-creased the activities of NOS in liver of piglets (P〈0.05). Supplementation of 1.6 % Arg significantly in-creased the activities of NOS in liver of piglets under oxidative stress (P〈0.05). 3) Oxidative stress signifi-cantly decreased IGF-Ⅰ, insulin-like growth factor-Ⅰ receptor ( IGF-ⅠR ) and insulin-like growth factor-binding protein 3 (IGFBP3) mRNA expression levels in liver of piglets (P〈0.05), but had no significant effects on the mRNA expression levels of those 3 genes in muscle (P〉0.05).
