摘 要:为研究禽网状内皮组织增殖病病毒(REV)对禽流感病毒(AIV)疫苗免疫效果的影响,本研究将1日龄SPF鸡经腹腔感染REV HLJR0901株后,分别于10日龄免疫重组AIV灭活疫苗(H5N1亚型,Re-6)、禽流感-新城疫重组二联活疫苗(r LH5-6)及10日龄和24日龄两次免疫r LH5-6疫苗,并设未感染REV的免疫对照组,于免疫后检测血清HI抗体,并采用H5N1亚型高致病性AIV攻毒。结果显示:实验鸡感染REV后HI抗体滴度均低于相应的未感染REV的免疫对照组。感染REV后,免疫一次Re-6灭活疫苗、r LH5-6活疫苗及两次r LH5-6活疫苗的实验鸡攻毒后的存活率分别为70%、60%及80%,排毒鸡数分别为4/10、4/10及3/10;未感染REV的免疫鸡均不发病、不死亡、不排毒;空白对照组实验鸡攻毒后3 d内全部死亡并且排毒。本研究表明,REV对AIV灭活疫苗和活疫苗均具有明显的免疫抑制作用,加强免疫r LH5-6疫苗可以部分减轻该抑制作用。To investigate the influence of reticuloendotheliosis virus (REV) infection on avian influenza (AI) vaccine, groups of SPF chickens were infected intraperitoneally with REV HLJR0901 strain at one day old, and vaccinated with inactivated H5N1 AI vaccine (Re-6) or live recombinant Newcastle disease virus H5 AI vaccine (rLH5-6) at 10 day-old or vaccinated with rLH5-6 twice at 10 days and boosted at 24 days of age, respectively, and followed by challenging with H5NI highly pathogenic avian influenza virus (HPA1V). The results showed that the titers of antibody against AIV detected by hemagglutination-inhibition test were significantly lower in the groups of REV infected chickens than that in the non-REV infected groups. Moreover, the immuno-protections of chickens vaccinated once with the inactivated Re-6, live rLH5-6 and twice with live rLH5-6 were 70%, 60% and 80%, respectively, and the virus shedding were 4/10, 4/10, and 3/10, respectively. However, the immunized chickens without REV infection were 100% protected without any clinical signs, deaths or virus shedding. While, all the chickens in blank control group died and shed virus within 3 days post-challenge with H5N1 HPAIV. Our study demonstrated that REV had a significant impact on both inactivated Re-6 and live rLH5-6 vaccines by inducing immuno-suppression in the chickens, and the boosting of rLH5-6 was able to slightly reduce the immunosuppressive effect of REV infection.
