太白银莲花皂苷6抑制U87细胞增殖并诱导凋亡

摘　　要：目的探讨太白银莲花皂苷6对U87胶质母细胞瘤细胞增殖的影响及可能机制。方法用(0.0、1.6、3.2、6.4、12.8)μg/m L太白银莲花皂苷6处理U87细胞24h、48h,采用MTT法检测太白银莲花皂苷6对细胞增殖的抑制作用;annexinⅤ-FITC/PI双染色结合流式细胞术检测细胞凋亡;Western blot法检测活化的caspase-3蛋白表达。结果与对照组相比,太白银莲花皂苷6能明显抑制U87细胞增殖,并且呈时间、剂量依赖性;随着药物浓度增加,U87细胞的凋亡率增加,活化caspase-3蛋白表达增强。结论太白银莲花皂苷6可呈剂量依赖性抑制U87细胞增殖,增加活化caspase-3蛋白表达,促进凋亡。Objective To investigate the effect of saponin 6 of Anemone Taipaiensis on the proliferation of human U87 MG glioma cells and the possible mechanism.Methods U87 MG cells were treated with different concentrations of saponin 6（0.0,1.6,3.2,6.4,12.8 μg/m L） for 24 hours or 48 hours.Cell viability was measured by MTT assay; the apoptosis rate was detected by flow cytometry combined with annexin V-FITC / PI staining; Western blotting was applied to determine the protein level of activated caspase-3.Results Compared with control groups,saponin 6 significantly inhibited U87 MG cell proliferation in a time-and dose-depended manner.Apoptosis rate of U87 MG cells and the expression of activated caspase-3were raised with the increasing concentration of saponin 6.Conclusion Saponin 6 of Anemone Taipaiensis could depress cel proliferation in a dose-depended manner,increase the expression of activated caspase-3 and promote apoptosis in U87 MG cel s.
