摘  要:目的:检测各理化因素对重组鸡痘病毒rFPVHg-Hp的影响,为临床前研究奠定基础。方法:将重组鸡痘病毒rFPVHg-Hp经热、酸、碱、乙醚、氯仿、胰蛋白酶、超声破碎、紫外线照射处理后,接种于CEF细胞,通过测定TCID50观察重组鸡痘病毒rFPVHg-Hp毒力活性的变化,由此分析这些理化因素对重组鸡痘病毒rFPVHg-Hp的影响。结果与结论:重组鸡痘病毒rFPVHg-Hp不耐高温,在55℃作用45 min或60℃作用15 min即可完全灭活;但是其对酸和乙醚具有一定的耐受性,在pH值为3-9的范围内,pH值的变化对病毒滴度无明显影响;同时紫外线照射无法使得培养基中的病毒粒子完全灭活;该病毒对氯仿、胰蛋白酶较为敏感,二者均可使病毒滴度下降;在适应的条件下,重组鸡痘病毒rFPVHg-Hp对超声波具有一定的耐受性。Objective: To detect the influence of rFPVHg-Hp which was disposed by physical and chemical factors and provide a foundation for the following clinical study. Methods: The CEF cells were infected by the rFPVHg-Hp which was disposed by heat, acid-base, ether, chloroform, trypsin, ultrasonic waves and ultraviolet rays, and then inspected the changes in virus titer to analyze the influence of rFPVHg-Hp which was disposed by these physical and chemical factors. Results Conclusion: The results showed that rFPVHg-Hp was sensitive to heat. It was completely inactivated by 55℃, 45 min or 60℃, 15 min. rFPVHg-Hp was tolerant of acid-base and ether, and the titer of rFPVHg-Hp was not significantly affected when the pH was changed in a range of 3-9. The virion in the medium was not completely inactivated by ultraviolet rays, and the titer of rFPVHg-Hp was reduced by chloroform and trypsin. rFPVHg-Hp was tolerant of ultrasonic waves in the right conditions.
