摘  要:目的:探讨血清微小RNA(mi RNAs)用于诊断子宫内膜异位症(内异症)的潜能。方法:利用分别混有10例内异症患者(内异症组)/对照组血清的样本,应用Taq Man micro RNA array方法对765种mi RNAs的表达情况进行检测;选择表达明显差异的mi RNAs分别在内异症组及对照组血清进行验证;进一步在内异症合并不孕、单纯内异症及输卵管性不孕三组患者中检测已验证的mi RNAs的表达。结果:在内异症组及对照组血清中存在6种mi RNAs(mi R-199a、mi R-122、mi R-145*、mi R-141*、mi R-542-3p及mi R-9*)的表达有明显差异;它们对内异症的诊断率较高,其中准确性最高的是mi R-145*,其AUC可高达0.883(95%CI:0.814-0.953,P<0.001)。此外,mi R-199a和mi R-122的表达在内异症合并不孕与输卵管性不孕的比较中有明显差异;mi R-199a和mi R-9*在内异症合并不孕与单纯性内异症有明显差异。结论:血清mi RNAs有望用于无创诊断内异症,其中mi R-199a有望用于预测内异症患者的生育能力。Objective : To explore the diagnostic utility of serum micro RNAs(mi RNAs) in endometriosis.Methods:765 serum mi RNAs were profiled using a Taq Man Micro RNA Array in a pool of 10 patients with endometriosis and a pool of 10 negative controls and a set of selected mi RNAs were further analyzed in a validation cohort. The validated mi RNAs were further detected in endometriosis complicated infertility, pure endometriosis and tubal infertility only. Results : The expression of 6 mi RNAs(mi R-199 a 、mi R-122 、mi R-145 * 、mi R-141 * 、mi R-542-3pand mi R-9*) were significantly different between endometriosis and controls. The diagnostic value of AUC were all higher than 0.8, and the highest was 0.883(95%CI :0.814-0.953,P〈0.001)of mi R-145 *. Furthermore, there were differential expression of mi R-199 a and mi R-122 between endometriosis complicated infertility and tubal infertility only, mi R-199 a and mi R-9 * between endometriosis complicated infertility and pure endometriosis. Conclusions :Serum mi RNAs may be a non-invasive biomarker for diagnosing endometriosis, and mi R-199 a may be used for predicting the fertility potential of endometriosis.
