摘 要:目的:探讨中药康肾合剂(KSP)对酒精性肾损害模型大鼠肾组织核转录因子κB(NF-κB)活化及白细胞介素6(IL-6)水平的影响。方法:将SD雄性大鼠66只随机分为正常组、酒精组、中药组各22只。正常组给予生理盐水灌胃,酒精组给予无水乙醇每日2次灌胃,中药组在给予同酒精组等量乙醇基础上,加用KSP水煎剂每日2次灌胃,共12周。于实验第4周、8周、I2周末采用免疫组化法及ELISA法检测肾组织NF-κB活化及IL-6水平变化。结果:酒精组大鼠肾组织NF-κB活化较正常组显著上调,IL-6水平明显上升(P<0.05);中药组NF-κB活化及IL-6水平明显低于酒精组(P<0.05)。结论:KSP干预酒精性肾损害的作用机制可能与其抑制NF-κB活化、降低IL-6水平有关。Objective: To study the effect of Kang Shen prescription(KSP) on the activation of nuclear factor kappa-B(NF-κB) and the level of Interleukin 6(IL-6) in rats of alcoholic renal damage. Methods: The 66 male SD rats were randomized into normal, alcoholic and Chinese medical groups with 22 cases in each. The normal salines were given to the rats of normal group by gavage, the ethanol for twice a day to the rats of alcoholic group and KSP based on the ethanol equal to the amount of the alcoholic group to the rats of Chinese medical group for twice day,with 12 weeks course. At the end of 4th, 8th and 12 th week, the NF-κB activation and IL-6 levels were determined by immunohistochemical and ELISA methods. Results: Compared with the normal group, the activation of NF-κB and the level of IL-6 in rats renal tissues of alcoholic group significantly went up(P〈 0.05); those in the Chinese medical group were markedly lower than in the alcoholic group(P〈 0.05). Conclusion: KSP intervention for patients with alcoholic kidney damage may be relevant to its functions in inhibiting the activation of NF-κB and decreasing the level of IL-6.
