摘 要:目的探讨脑心通胶囊对苯巴比妥成瘾大鼠的脑细胞损伤的保护作用并初步探讨其机制。方法用递增剂量的苯巴比妥诱导建立成瘾大鼠模型,并用脑心通胶囊进行干预,通过戒断症状评分、脑切片HE染色和免疫组化染色的方法探讨脑心通胶囊对苯巴比妥成瘾大鼠脑细胞损伤的保护作用及机制。结果苯巴比妥成瘾大鼠戒断症状评分较高,脑细胞受损伤而形态改变,脑额叶凋亡细胞数增多;用脑心通胶囊干预可以降低大鼠戒断症状评分,保护脑细胞形态完整,减少脑细胞凋亡,且脑心通胶囊对凋亡相关因子Bcl-2和Bax表达调控呈现浓度相关性。结论脑心通胶囊可以一定程度减轻苯巴比妥成瘾的戒断症状,保护脑细胞,其机制可能与凋亡调控因子Bcl-2和Bax有关。[ Objective ] To investigate the effect of Naoxintong on brain cells in phenobarbital-dependant and - withdrawn rats, and explore the related mechanisms. [ Methods ] The protective effect and mechanism of Naoxintong on the neurocytes were evaluated in phenobarbital-dependent rats. The withdrawal syndromes of rats were scored. The neurocytes were observed and the expressions of Bcl-2 and Bax protein of frontal cortex were examined after sacrificing the rats. [Results] The withdrawal syndromes scores of the phenobarbital-dependent rats were significantly different from those of the rest; Naoxintong-interventional rats expressed more Bcl-2 protein and less Bax protein in frontal cortex than phenobarbital-dependent rats. Naoxintong could keep the integrity of neurocytes and reduce brain cell apoptosis, and its regulation to Bcl-2 and Bax were concentration dependent. [ Conclusions ] Naoxintong could relieve the withdrawal syndromes and protect the nenrocytes from Phenobarbital. The mechanism of this effect may be related to the regulation of Bcl-2 and Bax.
