摘  要:目的:用基因芯片技术研究电磁脉冲(electromagnetic pulse,EMP)辐照对大鼠左心室肌组织的生物效应。方法:10只雄性同窝SD大鼠随机分为辐照组与正常对照组2组,每组5只。在锥形平板GTEM小室内对辐照组大鼠进行EMP辐照,EMP辐照参数为:场强200 k V·m-1,脉冲前沿3.5 ns,脉宽14 ns,重复频率1 Hz;正常对照组进行假辐照。辐照后18 h取大鼠左心室肌,提取RNA,经反转录后用Cy3、Cy5荧光标记,获得两组动物来源c DNA探针,c DNA探针与基因表达谱芯片进行杂交,结果由扫描仪扫描并用软件进行分析统计。结果:筛选出差异表达基因108条,表达增强的有51条,其中功能已知或部分已知的11条,占22%;表达减弱的有57条,其中功能已知或部分已知的15条,占26%。差异表达基因主要涉及代谢、神经递质、肌肉收缩、凋亡、应激等方面。其中蛋白激酶C、热休克蛋白70、辅酶Q10、亚甲基四氢叶酸脱氢酶、5-羟色胺受体2C、锌指蛋白10、干扰素γ、雄激素调节蛋白、雌激素调节蛋白等可能在EMP对大鼠心脏生物学效应方面起重要作用。结论:EMP辐照可诱导大鼠左心室肌组织多个基因特异性表达。Objective: To explore the bio-effects of electromagnetic pulse( EMP) on left ventricle myocardium of rats by the means of gene chip. Methods: 10 male SD rats were randomly divided into two groups with 5 for each group. One group was subjected to EMP fields( 200 k V·m^-1,3. 5 ns duration,14 ns width,1 Hz) and the other as control. 18 hours after the irradiation,the rats were sacrificed and their left ventricle myocardium were eviscerated. The fluorescent c DNA probes labeled with Cy3 and Cy5 were prepared from RNA extracted from the ventricle myocardium of the two groups. Probes of the two groups were then hybridized against c DNA gene chip,the fluorescent signals were scanned with a scanner and the results were analyzed by computer. Results: Compared with the control,108 genes in gene expression profile were altered. The expression levels of 51 genes were upregulated distinctly while 57 genes were down-regulated significantly involved in metabolism,neurotransmitter,muscle contraction,apoptosis,stress response and so on. Among the 108 genes,PKC,HSP70,ubiquinone oxidoreductase,methylene tetrahydrofolate dehydrogenase,5-hydroxytryptamine receptor 2C,zinc finger protein10,interferon gamma,androgen regulated 20 k Da protein,estrogen receptor associated protein and so on might have important roles on the cardiac biological effects by EMP. Conclusion: EMP exposure in this study can induce specific expressions of some genes in left ventricle myocardium of rats.
